摘 要:目的探究β-榄香烯注射液(ELE)联合甲磺酸阿帕替尼(APA)对乳腺癌细胞生长的抑制作用。方法正常培养乳腺癌细胞MB-468作空白对照组,用0. 05 mg/ml ELE单独或联合1μmol/L APA处理MB-468细胞,分别作ELE组、APA组和ELE+APA组。采用MTT法检测各组细胞生长情况,流式细胞仪检测细胞凋亡情况,Western blot检测Ki67、PCNA、Caspase-3、Bax、Bcl-2、Beclin1、p62、LC3的表达;用自噬抑制剂3-MA处理MB-468细胞,并进行ELE单独或联合APA干预,细胞分组为:对照组,3-MA组,ELE+APA组和3-MA+ELE+APA组,采用Western blot检测LC3的表达。结果与对照组相比,ELE组、APA组和ELE+APA组细胞增殖活性、Ki67、PCNA、p62的表达显著下调,凋亡率、Caspase-3、Bax/Bcl-2、Beclin1、LC3Ⅱ/LC3Ⅰ的表达显著上调;加入自噬抑制剂3-MA后,3-MA+ELE+APA组Beclin1、LC3Ⅱ/LC3Ⅰ的表达显著下调,p62的表达显著上调。结论 ELE联合APA对乳腺癌细胞MB-468生长有明显的抑制作用,可能与降低MB-468细胞增殖活性、促进其凋亡和自噬有关。Objective To investigate the inhibition effects of β-elemene(ELE) injection combined with apatinib mesylate(APA) on growth of breast cancer cells. Methods The breast cancer cells MB-468 were normally cultured and enrolled as blank control group(Control).MB-468 cells treated with 0. 05 mg/m LELE injection,1 μmol/L APA or their combination were included in ELE group,APA group and ELE with APA group,respectively. The cells growth of all groups was detected by MTT assay. Apoptosis was detected by flow cytometry. Expression of Ki67,PCNA,Caspase-3,Bax,Bcl-2,Beclin1,p62 and LC3 was detected by Western blot. MB-468 cells were treated with autophagy inhibitor 3-MA,The intervention with ELE alone or its combination with APA was performed. The cells were divided into Control group,3-MA group,ELE with APA group and 3-MA with ELE and APA group. The expression of LC3 was detected by Western blot. Results Compared with control group,cell proliferation activity,expression of Ki67,PCNA and p62 were significantly down-regulated in ELE group,APA group,ELE with APA group,while apoptosis rate,expression of Caspase-3,Bax/Bcl-2,Beclin1 and LC3Ⅱ/LC3Ⅰ were significantly up-regulated.After addition of autophagy inhibitor 3-MA,expression of Beclin1 and LC3Ⅱ/LC3Ⅰ in 3-MA with ELE and APA group was significantly down-regulated,while expression of p62 was significantly up-regulated. Conclusion There are significant inhibition effects of ELE injection combined with APA on growth of breast cancer cells MB-468,which may be related to decreasing MB-468 cell proliferation activity,promoting apoptosis and autophagy.
