摘　　要：目的研究热休克蛋白90(Heat Shock Proteins 90,HSP90)分子伴侣体系,作为抗肿瘤靶标,在药物化学中的研究新进展,为其深入研究与应用提供参考。方法参考国内外70余篇新近研究成果,从HSP90的生物结构、功能、特征及其抑制剂等方面进行简要概述。结果HSP90由于其在肿瘤的发生和发展中所起到的重要作用,已经越来越受到肿瘤药理学家的关注。近年来发表的与HSP90相关的研究论文愈来愈多。从已发表论文的研究结果中,可看出以HSP90为抗肿瘤靶标开发HSP90抑制剂,已广泛被药物化学家们所认同,并成为当前抗肿瘤药物研发的热点之一。结论HSP90抑制剂临床表现治疗效果较好,但更进一步优化临床给药方案、寻找高效安全的组合用药治疗方法、个性化精准医治则是未来HSP90抑制剂研究的新方向,新突破可能就在于此。Purposes—To study the new research progress of heat shock protein 90(HSP90)molecular chaperone system as a potential antitumor target in medicinal chemistry,and to provide reference for its further research and application.Methods—With more than 70 research results both at home and abroad as reference,the new progress of related HSP90 research is summarized from its biological structure,function,characteristics and its inhibitors.Result—More and more attention has been paid to the important role of HSP90 in the occurrence and development of tumor by tumor pharmacologists.The published papers related to HSP90 have been increased sharply in recent years.From the results of these papers,medicinal chemists have widely recognized the HSP90 inhibitors which are developed with HSP90 as antitumor target,and the HSP90 inhibitors has become one of the hotspots in the research and development of antitumor drugs.Conclusion—Although HSP90 inhibitors have shown good effect in clinical treatment,further optimization of drug administration in clinical trials,the design of high-efficient and safe combination therapy method,individual precision medical treatment will be the next research direction of HSP90 inhibitors,a new breakthrough may be broken in this aspect.
