摘 要:目的通过观察芪红水煎液对免疫性肝纤维化大鼠肝组织血小板衍生因子(PDGF)信号通路关键蛋白的影响并探讨其抗肝纤维化的作用机制。方法 SD大鼠随机分为正常对照组、模型组、芪红水煎液组和秋水仙碱组,每组7只。除外正常对照组,其他所有各组均采用猪血清腹腔注射诱导并建立免疫性肝纤维化模型。模型成功后,正常对照组和模型组给以等量0.9%氯化钠注射液,芪红水煎液组和秋水仙碱组灌服相应药物6周。光镜下观察大鼠肝脏病理学HE染色;血清生化法检测大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(GOT)、碱性磷酸酶(ALP)含量;碱水解法检测肝脏羟脯氨酸(Hyp)含量;Western blotting法观察肝脏PDGFBB、血小板衍生因子受体(PDGFR)蛋白的表达。结果猪血清诱导的肝纤维化大鼠肝组织病理有明显的胶原纤维增生,肝功能指标水平明显升高,肝组织中羟脯氨酸含量明显增加,芪红水煎液在一定水平上改善肝纤维化病理形态,减低肝脏PDGF-BB、PDGFR蛋白的含量,提示芪红水煎液可通过抑制PDGF信号通路介导的肝星状细胞活化、增殖来抵抗肝纤维化。结论芪红水煎液可能抑制由PDGF信号通路激活的HSC来有效拮抗猪血清诱导的大鼠免疫性肝纤维化。Objective:To observe the effect of Qihong Decoction on the key protein of PDGF signaling pathway in liver tissue of rats with immune hepatic fibrosis and explore its anti-fibrosis mechanism.Methods:SD rats were randomly divided into the normal control group,the model group,Qihong Decoction group and colchicine group,7 cases in each.Except the normal control group,other groups were induced by intraperitoneal injection of porcine serum to establish an immunological liver fibrosis model.After the model was successful,the normal control group and the model group were given the same amount of normal saline;Qihong Decoction group and colchicine group were administered with the corresponding drugs for 6 weeks.Liver pathological HE staining was observed under light microscope;serum ALT,GOT and ALP contents were detected with serum biochemical method;liver Hyp content was detected with alkali hydrolysis method;liver PDGF-BB and PDGFR protein expression was observed with Western blotting method.Results:Liver tissue pathology of liver fibrosis induced by pig serum had obvious collagen fibrosis;liver function index level was significantly increased,and hydroxyproline content in liver tissue was significantly increased.Qihong Decoction could improve the pathological morphology of liver fibrosis at a certain level,protect liver function,and reduce the content of PDGF-BB and PDGFR protein in liver,suggesting that Qihong Decoction could resist liver fibrosis by inhibiting the activation and proliferation of hepatic stellate cells mediated by PDGF signaling pathway.Conclusion:Qihong Decoction may inhibit HSC activated by PDGF signaling pathway to effectively antagonize pig serum induced immune liver fibrosis in rats.
