摘 要:目的检测微小RNA-186(miR-186)对非小细胞肺癌(non-small-cell lung cancer,NSCLC)细胞的影响,组织中的表达及其与临床特征的相关性分析。方法实时荧光定量聚合酶链反应(qRT-PCR)检测NSCLC组织、NSCLC旁组织、NSCLC细胞和人正常肺上皮细胞中miR-186的表达水平;miR-186高表达组NSCLC患者与低表达组患者的5年生存率,采用Log-rank分析;MTT实验检测细胞的增殖;Transwell实验检测细胞的侵袭能力;卡方检验分析miR-186与NSCLC患者临床特征的相关性。结果NSCLC组织中miR-186的表达水平低于NSCLC旁组织;NSCLC细胞系中miR-186的表达水平低于人正常肺上皮细胞;miR-186低表达的NSCLC患者5年生存率低于miR-186高表达组;miR-186抑制NSCLC细胞的增殖和侵袭;miR-186的表达水平与NSCLC患者的TNM分期、病理分级和淋巴结转移相关。结论miR-186可抑制NSCLC细胞的增殖和侵袭,在NSCLC组织中低表达与患者的TNM分期、病理分级和淋巴结转移相关,并影响患者的生存率。Objective To detect the effect of miR-186 on non-small-cell lung cancer(NSCLC)cells,the expression in tissues and its correlation with clinical features.Methods Real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression level of miR-186 in NSCLC tissues,NSCLC parasites,NSCLC cells and human normal lung epithelial cells.The 5-year survival rate of patients with NSCLC in the miR-186 high and low expression group were analyzed by Log-rank analysis.MTT assay was used to detect cell proliferation.Transwell assay was used to detect cell invasion ability.Chi-square test was used to analyze miR-186 and NSCLC patients clinical features.Results The expression level of miR-186 in NSCLC tissues was lower than organization next to NSCLC tissues.The 5-year survival rate of patients with miR-186 low expression was lower than that of miR-186 high expression.miR-186 inhibited proliferation and invasion of NSCLC cells.The expression level of miR-186 was associated with TNM staging pathological grade and lymph node metastasis in NSCLC patients.Conclusion miR-186 can inhibit the proliferation and invasion of NSCLC cells and is low in NSCLC tissues,which is associated with TNM staging,pathological grade and lymph node metastasis.
