摘 要:目的分析氯化锂(LiCl)对大鼠脑缺血再灌注损伤机制及血胰岛素水平的影响。方法选取SD大鼠54只,随机分为3组,假手术组、缺血组(脑缺血造模)、LiCl处理组,每组18只。分析3组大鼠脑缺血再灌注、神经功能、血浆胰岛素水平情况。结果实验动物脑缺血再灌注后24 h,3组实验动物的脑梗死体积、凋亡细胞数比较,差异有统计学意义(P<0.05),其中脑缺血组的以上指标均高于LiCl处理组、假手术组,且LiCl处理组均高于假手术组(P<0.05)。实验动物脑缺血再灌注后24 h,3组实验动物的神经功能缺损评分比较,差异有统计学意义(P<0.05),其中脑缺血组(2.41±0.43)分明显高于LiCl处理组的(1.34±0.49)分、假手术组0分,且LiCl处理组明显高于假手术组(P<0.05)。缺血30 min、缺血1 h时,脑缺血组与LiCl处理组的血浆胰岛素水平明显低于术前与假手术组(P<0.05),而再灌注后6 h、12 h及24 h,脑缺血组的血浆胰岛素水平均明显低于LiCl处理组与假手术组(P<0.05)。结论脑缺血再灌注可降低血浆胰岛素水平,而LiCl处理作用PI3K-PKB/Akt-GSK-3β信号通路可降低脑缺血再灌注脑梗死体积、细胞数凋亡数及神经功能缺损,提高血浆胰岛素水平,有一定修复脑组织损伤的效果。Objective To investigate the effects of lithium chloride(LiCl) on cerebral ischemia reperfusion injury and plasma insulin levels in rats.Methods Fifty-four SD rats were randomly divided into three groups:sham operation group,cerebral ischemia group(cerebral ischemia model) and LiCl treatment group,with 18 rats in each group.The brain ischemia-reperfusion,neurological function and plasma insulin levels were observed and compared among the three groups.Results At 24 hours after cerebral ischemia reperfusion, there were significantly differences in the cerebral infarction volume and number of apoptotic cells among the three groups(P<0.05).The above indexes in cerebral ischemia group were significantly higher than LiCl treatment group and sham operation group,moreover,which in LiCl treatment group were significantly higher than those in sham operation group(P<0.05).At 24 hours after cerebral ischemia reperfusion, there were significantly differences in the neurological deficit scores among the three groups(P<0.05),which in cerebral ischemia group [(2.41±0.43)points] were significantly higher than those [(1.34±0.49)points] in LiCl group and those(0 point) in sham operation group,which in LiCl treatment group were significantly higher than those in sham operation group(P<0.05).At 30 min and 1 h after cerebral ischemia,the plasma insulin levels in cerebral ischemia group and LiCl group were significantly lower than those before operationj and those in sham groups(P<0.05),and at 6 h,12 h and 24 after reperfusion, the plasma insulin levels in cerebral ischemia group were significantly lower than those in LiCl treatment group and sham operation group(P<0.05).Conclusion The cerebral ischemia-reperfusion can reduce plasma insulin levels in rat models,and LiCl treatment can reduce the cerebral infarct volume,number of apoptotic cells and neurological deficits,and increase plasma insulin levels after cerebral ischemia-reperfusion injury by means of PI3 K-PKB/Akt-GSK-3β signaling pathway,which has a certain effect of repairi
