摘  要:目的研究正畸牙移动(OTM)过程中外周神经对Wnt信号通路的调控以及它们在正畸成骨过程中的作用。方法分别建立OTM模型和辣椒素去感觉神经模型:将36只SD大鼠随机分为不注射辣椒素+不进行牙移动组(对照组)、不注射辣椒素+牙移动组(牙移动组)、注射辣椒素+不进行牙移动(去神经组)、注射辣椒素+牙移动(去神经+牙移动组),每组各9只大鼠。使用逆转录定量PCR(qRT-PCR)检测移动牙周围牙槽骨Wnt5a和Wnt10b的mRNA表达水平,通过免疫组织化学染色(免疫组化)检测各组右侧上颌第一磨牙牙根张力侧Wnt5a和骨钙素的表达变化。结果在SD大鼠上进行OTM后,牙槽骨中Wnt5a和Wnt10b mRNA水平上调(P均<0.05)。使用辣椒素去感觉神经后,有OTM大鼠Wnt5a mRNA与无OTM大鼠相近(P>0.05),而Wnt10b mRNA水平仍升高(P<0.05)。免疫组化示,正常SD大鼠OTM 9 d后,张力侧牙周膜内Wnt5a蛋白和成骨标志物骨钙素蛋白表达水平上调(P均<0.05),但辣椒素去感觉神经后Wnt5a和骨钙素蛋白表达水平被显著抑制,与无OTM大鼠相近(P均>0.05)。结论感觉神经在OTM诱导的骨改建过程中起着重要的作用。感觉神经可能通过调控Wnt5a的表达而调控OTM中的骨改建过程。Objective To investigate whether peripheral sensory nerves regulate the Wnt signaling pathway and their potential roles in the bone formation during the process of orthodontic tooth movement(OTM).Methods SD rat models with OTM and sensory denervation with capsicin were established.Thirtysix SD rats were randomly and evenly divided into four groups:no capsaicin injection+no OTM group(control group),no capsaicin injection+OTM group(OTM group),capsaicin injection+no OTM group(denervation group)and capsaicin injection+OTM group(denervation+OTM group).The expression levels of Wnt5a and Wnt10b mRNA in the alveolar bone were measured by qRT-PCR.The changes in the expression of Wnt5a and osteocalcin on the root tension side of the right maxillary first molar were measured by immunohistochemistry.Results The expression levels of Wnt5a and Wnt10b mRNA were significantly up-regulated after OTM in SD rats(both P<0.05).After sensory denervation with capsaicin,the expression of Wnt5a mRNA in SD rats with OTM was similar to that in SD rats without OTM(P>0.05),whereas the expression level of Wnt10b mRNA was significantly up-regulated(P<0.05).Immunohistochemistry revealed that the expression levels of Wnt5a and osteocalcin(osteogenic marker)proteins on the tension side were significantly up-regulated in normal SD rats at 9 d after OTM(both P<0.05),whereas remarkably down-regulated after sensory denervation with capsicin,which were similar to that in SD rats without OTM(both P>0.05).Conclusion Sensory nerves play a pivotal role in the OTM-induced bone formation,which probably modulate the bone formation during OTM by regulating the expression level of Wnt5a.
