摘  要:目的探讨二十二碳六烯酸(DHA)对由孕期抑郁引起的子代抑郁行为和认知能力的影响以及其可能的机制。方法通过长期束缚构建抑郁孕鼠模型。强迫游泳和悬尾实验用以检测小鼠抑郁状态,新物体和新位置识别实验用以检测小鼠的认知能力;免疫组化方法检测小鼠海马神经干细胞的生长情况。结果相较于对照组小鼠,束缚应激组的小鼠在强迫游泳和悬尾实验中不动时间增加(P<0.001);对新物体和新位置的物体探索时间降低(P<0.001);以及成年海马神经发生数目减少(P<0.001)。孕鼠在束缚应激期补充DHA后子鼠相较于没有补充DHA孕鼠的子鼠,在强迫游泳和悬尾实验中不动时间减少(P<0.001);对于新物体和新位置的物体的探索时间增加(P<0.001);成年海马神经发生数目增加(P<0.001)。结论孕期补充DHA可以降低母鼠孕期应激引起的子鼠抑郁状行为以及认知损伤,其机制可能和增加子鼠的成年海马神经发生有关。Objective To investigate the effect of docosahexaenoic acid(DHA)on the depression behaviors and cognitive ability of the offspring induced by stress during pregnant and its underlying mechanism.Methods The model of depressed pregnant mice was established by long-term restraint.Forced swimming and tail suspension tests were used to detect depression in mice.Novel object and novel location tests were used to observe the cognitive ability of mice.The adult hippocampal neurogenesis was measured by immunohistochemistry.Results Compared offspring mice of depressed pregnant mice with offspring mice of normal pregnant mice,we found that the immobility time of the depressed mice in the forced swimming(P<0.001)and tail suspension experiment(P<0.001)increased compared with the normal mice;the time for the exploration of the new objects(P<0.001)and the new positions(P<0.001)was decreased;and the adult hippocampal neurogenesis decrease(P<0.001).Compared offspring mice of depressed pregnant mice with DHA supplementation with offspring mice of depressed pregnant mice without DHA supplementation,we found that the time of the DHA mice in the forced swimming(P<0.001)and tail suspension experiment(P<0.001)decreased compared with the mice without DHA,the time of the DHA mice for the exploration of the new objects(P<0.001)and the new positions(P<0.001)was increased,and the adult hippocampal neurogenesis of the DHA mice was similar to that of the normal mice(P<0.001).Conclusion Supplementation of DHA during pregnancy can ameliorate depressive-like behavior and cognitive impairment of offspring induced by perinatal stress,which may be related to the increase of adult hippocampal neurogenesis in offspring.
