摘 要:目的探究吗啡成瘾小鼠的大脑前额叶皮层中lncRNA表达谱的变化。方法将C57BL/6小鼠分为两组:吗啡组和对照组。吗啡组小鼠以剂量按日递增的方法腹腔注射盐酸吗啡,建立急性吗啡成瘾的小鼠模型。对照组小鼠每日注射与吗啡组小鼠相同体积的生理盐水。提取两组小鼠大脑前额叶皮层(PFC)的总RNA,利用lncRNA表达谱芯片检测PFC中lncRNA的表达量变化。从芯片检测结果中筛选出表达量发生显著变化,且差异具有统计学意义的前10个lncRNA,并用qRT-PCR的方法验证其表达量变化。结果芯片结果显示,有493个lncRNA表达上调,255个lncRNA表达下调。qRT-PCR的验证结果表明,与对照组相比,吗啡组小鼠的大脑前额叶皮层中有5个lncRNA表达量发生显著变化,差异具有统计学意义(P<0.01),其中AK157323、AK018061和AK140599表达显著上调,AK007908和AK160450表达显著下调。结论吗啡成瘾可诱导小鼠大脑前额叶皮层中lncRNA表达谱发生较大变化。AK157323、AK007908、AK160450、AK018061和AK140599的表达异常可能参与吗啡成瘾的过程。Objective To explore the alterations of the lncRNA expression profile in the prefrontal cortex(PFC)of the morphine-addicted mouse.Methods C57BL/6 mice were divided into two groups:morphine group and control group.Mice in morphine group were intraperitoneally injected with morphine hydrochloride with daily increasing dose to establish the acute morphine-addicted model.Mice in control group were daily given the same volume of saline injection as those in morphine group.The total RNA of PFC was extracted to analyze the lncRNA expression profile by the lncRNA expression profile microarray.Then,the top 10 lnRNAs with significant changes in expression level and statistically significant difference were screened from the lncRNA microarray results,and verified subsequently by the qRT-PCR.Results The results of lncRNA microarray displayed 493 up-regulated lncRNA and 255 down-regulated lncRNA.The verification results of qRT-PCR showed that compared with control group,the expression levels of 5 lncRNAs in the PFC showed significant changes in morphine group(P<0.01).Among them,the expression of AK157323,AK018061 and AK140599 was significantly up-regulated,while the expression of AK007908 and AK160450 was significantly down-regulated.Conclusion Morphine addiction may induce the significant changes of lncRNA expression profile in the PFC of mice.The abnormal expression of AK157323,AK007908,AK160450,AK018061 and AK140599 may be involved in the process of addition induced by morphine.
