摘 要:目的观察复方苦参注射液对肺癌H1299细胞放射敏感度的影响,并探讨其作用机制。方法采用四甲基偶氮唑盐法检测放射对H1299细胞增殖的影响;采用流式细胞法检测H1299细胞凋亡率;克隆形成试验检测各组H1299细胞的存活率。结果不同浓度复方苦参注射液处理H1299细胞24、48、72 h的IC50分别为14.46、8.51、3.52 mg/mL,对H1299细胞的毒性有一定的剂量和时间依赖性。苦参放射组H1299细胞增殖抑制率分别为33.03%、34.00%,单纯放射组抑制率分别为12.73%、15.44%,差异有统计学意义(P<0.05)。苦参放射组H1299细胞的凋亡率为11.0%,单纯放射组凋亡率为6.4%,差异有统计学意义(P<0.05)。单纯放射组H1299细胞的存活率为16.3%,复方苦参注射液联合放射可使H1299细胞存活率降为12.0%,差异具有统计学意义(P<0.05)。结论复方苦参注射液能增加放射线诱导的肺癌H1299细胞凋亡,从而提高肺癌H1299细胞的放射敏感度。Objective To investigate the effect of compound sophora flavescens injection on radiation sensitivity of H1299 lung cancer cells and its underlying mechanism.Methods Methylthiazolyl tetrazolium(MTT)assay was used to observe the effect of compound sophora flavescens injection on the proliferation of H1299 cells after radiation and flow cytometry mothod to detect apoptosis rate of H1299 cells.The survival rate of cells in various groups was detected by clone formation assay.Results Different concentrations of compound sophora flavescens injection had cytotoxicity on H1299 cells.The IC50 for 24,48,and 72 h was 14.46 mg/mL,8.51 mg/mL,and 3.52 mg/mL respectively,in obvious dose and time dependent manner.The inhibition rate of proliferation was more significantly increased than that of the blank control group(P<0.05).The inhibition rates of proliferation of H1299 cells in compound sophora flavescens injection group were 33.03%and 34.00%respectively,and those in radiation group were 12.73%and 15.44%,respectively,with statistical significance(P<0.05).The apoptotic rate of H1299 cells in compound sophora flavescens injection group was 11.0%;that of radiation group 6.4%and the difference was statistically significant(P<0.05).The survival rate of H1299 cells in radiation group was 16.3%.Compound sophora flavescens injection combined with radiation could reduce the survival rate of H1299 cells to 12.0%and the difference was statistically significant(P<0.05).Conclusion Compound sophora flavescens injec tion can increase the apoptosis of lung cancer H1299 cells induced by radiation,thus enhance the radiosensitivity of H1299 lung cancer cells.
