摘 要:目的探究珠子参汤剂对脑胶质瘤U87细胞增殖和凋亡的影响,并对其分子机制进行初步探究。方法将脑胶质瘤U87细胞随机分为空白组(A组)、珠子参汤剂低剂量组(B组)、珠子参汤剂中剂量组(C组)、珠子参汤剂高剂量组(D组)。用MTT检测细胞增殖情况,流式细胞仪和TUNEL染色检测细胞凋亡情况,Real-time PCR检测细胞内Bcl-2 mRNA、Bax mRNA和Caspase-3 mRNA水平,Western blotting检测细胞内p-PI3K蛋白、p-AKT蛋白的水平。结果与A组比较,C、D组细胞存活率显著降低(P<0.05);B、C、D组Bcl-2 mRNA表达水平显著降低,B、C组Bax mRNA与Caspase-3 mRNA水平显著升高(P<0.05);B、C、D组p-PI3K蛋白与p-AKT蛋白表达水平显著降低(P<0.05)。与B组比较,C、D组细胞存活率显著降低(P<0.05);D组Bcl-2 mRNA表达水平显著降低,Bax mRNA与Caspase-3 mRNA水平显著升高(P<0.05);p-PI3K蛋白与p-AKT蛋白表达水平显著降低(P<0.05);与C组比较,D组细胞凋亡率、Bax mRNA水平显著升高,而Bcl-2 mRNA表达水平、p-AKT蛋白表达水平显著降低(P<0.05)。结论珠子参汤剂可以抑制脑胶质瘤U87细胞增殖、细胞凋亡,其机制可能与珠子参汤剂调控PI3K/AKT信号通路有关。Objective To investigate the effects of zhuzishen decoction on the proliferation and apoptosis of glioma U87 cells,and to explore its molecular mechanism.Methods U87 glioma cells were randomly divided into blank group(group A),low-dose group(group B),medium-dose group(group C)and high-dose group(group D).Cell proliferation was detected by MTT,apoptosis detected by flow cytometry and TUNEL staining,and intracellular levels of Bcl-2,Bax and C aspase-3 were detected by Real-time PCR.The levels of p-PI3 K and p-AKT protein were detected by Western blotting.Results Compared with group A,cell viability was significantly lower in group C and D(P<0.05);apoptosis significantly increased in group B,C and D;expression of Bcl-2 was significantly decreased in group B,C and D;expression of Bax and C aspase-3 significantly increased in group B and C(P<0.05);expression of p-PI3 K and p-AKT protein significantly decreased in group B,C and D(P<0.05).Compared with group B,the cell survival rate was significantly lower in group C and D(P<0.05);the apoptotic rate was significantly higher in group C and D(P<0.05);the expression level of Bcl-2 and Bax and C aspase-3 were significantly lower in group D(P<0.05);the expression levels of p-PI3 K and p-AKT were significantly lower(P<0.05);compared with group C,the apoptotic rate and Bax gene level were significantly higher in group D.However,the expression of Bcl-2 and p-AKT decreased significantly(P<0.05).Conclusion Zhuzishendecoction can inhibit the proliferation and apoptosis of glioma U87 cells,and its mechanism may be related to the regulation of PI3 K/AKT signaling pathway by Zhuzishen Decoction.
