摘 要:目的:探讨鸦胆子素D对人乳腺癌细胞株MDA-MB-231增殖、凋亡及细胞周期的影响。方法:MDA-MB-231细胞加入不同浓度的鸦胆子素D(0、3、5、10、30、50μmol/L)分别作用24 h、48 h、72 h后,CCK-8检测细胞增殖情况。流式细胞仪检测细胞凋亡及周期情况。结果:CCK-8结果提示经不同浓度鸦胆子素D作用后,MDA-MB-231细胞存活率下降,与对照组相比差异有统计学意义(P<0.05)。细胞凋亡结果提示经0、10、30、50μmol/L的鸦胆子素D作用24 h后,总细胞群中凋亡细胞百分比分别为6.69%、15.12%、35.83%和40.67%,与对照组差异有统计学意义(P<0.05)。细胞周期结果提示经0、10、30、50μmol/L的鸦胆子素D作用24 h后,G 0/G 1期乳腺癌细胞占比分别为31.45%、40.93%、57.16%、66.38%;S期乳腺癌细胞占比分别为40.12%、33.08%、29.32%、20.45%;G 2/M期乳腺癌细胞占比分别为28.43%、25.99%、13.52%、11.17%,与对照组相比差异有统计学意义(P<0.05)。结论:MDA-MB-231细胞存活率随鸦胆子素D药物浓度增加而降低,且时间越长细胞存活率越低,呈时间剂量依赖性。鸦胆子素D促进MDA-MB-231细胞凋亡比例增加且其增殖周期阻滞在C 0/G 1期。Objective:Investigate the effects of Bruceine D on proliferation,apoptosis and cell cycle of human breast cancer cell line MDA-MB-231.Methods:CCK-8 was used to detect the proliferation of Bruceine D(0,3,5,10,30,50)μmol/L MDA-MB-231 cell after 24 h,48 h,72 h,respectively.Apoptosis and cell cycle were detected by flow cytometry.Results:CCK-8 suggested that after Bruceine D at different concentrations,the survival rate of MDA-MB-231 cells decreased,and the difference was statistically significant compared with the control group(P<0.05).The results of apoptosis suggested that the percentage of apoptotic cells in the total cell population was 6.69%,15.12%,35.83%and 40.67%,respectively,after 24 h of Bruceine D treatment with 0,10,30 and 50μmol/L,which was statistically significant different from the control group(P<0.05).The cell cycle results suggested that after 24 h of Bruceine D treatment with 0,10,30 and 50μmol/L,the proportion of G 0/G 1 breast cancer cells was 31.45%,40.93%,57.16%and 66.38%;the proportion of S breast cancer cells was 40.12%、33.08%、29.32%and 20.45%;the proportion of G 2/M breast cancer cells was 28.43%、25.99%、13.52%and 11.17%,respectively,and the difference was statistically significant compared with the control group(P<0.05).Conclusion:The survival rate of MDA-MB-231 cells decreases with the increase of Bruceine D concentration,and the cell survival rate decreases with the increase of time.Bruceine D increased the apoptosis rate of MDA-MB-231 cells and its proliferation cycle is blocked at C 0/G 1 phase.
