摘 要:【目的】对马铃薯体细胞胚胎发生类受体激酶(somatic embryogenesis receptor-like kinases,SERKs)基因进行克隆,通过序列分析和表达分析,初步探究马铃薯体细胞胚胎发生类受体激酶是否能够作为共受体在植物免疫防御应答中发挥功能.【方法】以拟南芥SERKs家族基因氨基酸序列同源比对马铃薯基因组数据库,找到了3个马铃薯SERKs家族基因.利用高效的Gateway技术,从马铃薯中克隆了StSERK1,StSERK3A和StSERK3B 3个马铃薯SERKs家族基因;以拟南芥免疫激活型突变体bak1-3 bkk1-1为背景,通过遗传学、分子生物学及生理生化的方法对3个马铃薯SERKs基因在植物免疫调控中的作用进行基因功能研究.【结果】结构域分析和遗传进化树结果证实,StSERK1、StSERK3A和StSERK3B均具有体细胞胚胎发生类受体激酶结构域特征.组织表达分析显示,马铃薯3个SERKs基因均能在植物中表达.遗传数据证实,3个马铃薯SERKs基因均能不同程度抑制bak1-3 bkk1-1免疫激活的表型.【结论】马铃薯StSERKs基因具有SERKs家族基因的典型保守结构域特征,能够参与植物先天性免疫应答.【Objective】 The Solanum tuberosum somatic embryogenesis receptor-like kinases(SERKs) genes were cloned and whether those genes can act as a co-receptor in plant immunity response were studied through analysis on the gene sequences and gene expression. 【Methods】 Three potato SERKs family genes were identified in the potato genome database based on the amino acid sequence homolog of the SERKs family of Arabidopsis. By using the efficient Gateway cloning technology,three potato SERKs genes(StSERK1,StSERK3 A and StSERK3 B) were cloned from potato. The Arabidopsis immune-activated mutant bak1-3 bkk1-1 was used as background material to investigate the biological functions in potato immune. 【Results】 The results of gene domain analysis and genetic phylogenetic tree confirmed that StSERK1,StSERK3 A and StSERK3 B all contained the domain structural characteristics of SERKs family domain.Tissue expression analysis showed that three SERKs genes of potato could be expressed in plant,but the expression of different tissues varied. Genetic data confirmed that the three potato SERKs genes could inhibit the auto-immune phenotype of bak1-3 bkk1-1 in Arabidopsis. 【Conclusion】 Potato SERKs gene has typical conserved domain characterisics of SERKs family genes and can participate in plant innate immune response.
