摘 要:目的:探究神经菌毛素-1(NRP-1)对急性淋巴细胞白血病CCRF-CEM细胞的生物学特征的影响。方法:分别采用以下两种方法(构建NRP-1敲低CCRF-CEM细胞株或使用NRP-1单克隆抗体)抑制NRP-1功能,通过对比抑制NRP-1功能后CCRF-CEM细胞株与正常CCRF-CEM细胞株,确定NRP-1对CCRF-CEM细胞的生物学特性的影响。结果:成功构建NRP-1敲低CCRF-CEM细胞株(NRP-1-shRNA)。两种方法抑制NRP-1功能后,细胞的增殖、迁移能力均显著降低,而细胞凋亡及对化疗药物的敏感性增加。结论:NRP-1可促进急性淋巴细胞白血病CCRF-CEM细胞增殖和细胞迁移能力,抑制细胞的凋亡和降低细胞的化疗敏感性。Objective:To investigate the effect of neuropilin-1(NRP-1) on the biological characteristics of acute lymphoblastic leukemia CCRF-CEM cells.Methods: NRP-1 function was inhibited by the following two methods(viral transfection method to obtain NRP-1 knockdown of CCRF-CEM cell line or use of NRP-1 monoclonal antibody),and CCRF-CEM was inhibited by contrastive inhibition of NRP-1 function.The cell line was compared with the normal CCRF-CEM cell line to determine the effect of NRP-1 on the biological characteristics of CCRF-CEM cells.Results: The NRF-1 knockdown CCRF-CEM cell line was successfully constructed.After inhibiting the function of NRP-1 by both methods,the proliferation and migration of cells were significantly decreased,and apoptosis and sensitivity to chemotherapeutic drugs were increased.Conclusion: NRP-1 can promote the proliferation and migration of acute lymphoblastic leukemia CCRF-CEM cells and inhibit the apoptosis of cells and the sensitivity of chemotherapy.
