摘  要:长白山猪苓和陕西猪苓因地理隔离存在种间差异,为阐明长白山猪苓与陕西猪苓菌丝体间的基因表达特征,利用Illumina测序技术对长白山猪苓与陕西猪苓菌丝体的6份样品进行转录组测序,并对2种菌丝体的差异表达基因进行了比较分析。通过对转录组原始数据进行数据质控共得到133 662个Transcript和20 999个Unigene。差异表达分析结果表明:与陕西猪苓菌丝体相比,长白山猪苓菌丝体共有5 881个表达差异基因出现,其中,上调基因有2 616个,下调基因有3 265个。差异基因GO注释分类表明,在细胞组分中,差异表达基因主要分布在膜、膜部分、细胞、细胞部分和细胞器;分子功能中,差异表达基因主要与催化活性、结合有关;而在生物学过程中,主要与代谢过程和细胞过程有关。差异表达基因KOG注释分类表明,差异表达基因主要与一般功能基因预测、蛋白质翻译后修饰与转运、伴侣蛋白,次生代谢产物的生物合成、转运和分解代谢,信号转导机制,脂质运输和新陈代谢,氨基酸转运和代谢等有关。差异表达基因KEGG分类表明,差异表达基因主要涉及到碳水化合物代谢,翻译,氨基酸代谢,脂质代谢,运输和分解代谢,信号转导,折叠、分拣和降解等。There are interspecific differences between Changbaishan Polyporusumbellatus and Shaanxi Polyporusumbellatus due to geographical isolation. In order to understand the gene expression characteristics and provenance differences between the hyphae of the ChangbaishanPolyporusumbellatus and Shaanxi Polyporusumbellatus, six samples of Changbaishan and Shaanxi Polyporusumbellatus mycelium were sequenced, and the differentially expressed genes of the two myceliums were analyzed. As a result, a total of 133 662 transcripts and 20 999 unigenes were obtained by data quality control of the transcriptome raw data. Differential expression analysis indicated that there were 5 881 differentially expressed genes in the mycelium of Changbaishan Polyporusumbellatus compared with the mycelium of Shaanxi Polyporusumbellatus. Of which, there were 2 616 up-regulated genes, 3 265 down-regulated genes. The GO annotation classification of differentially expressed genes showed that the differentially expressed genes were mainly related with the membrane, membrane part, cell, cell part and organelle in the cell component. Among the differentially expressed genes, some genes were mainly related to catalytic activity and binding in the molecular function, the others were mainly related to metabolic process and cellular process in the biological process. The KOG annotation classification of differentially expressed genes indicated that differentially expressed genes are mainly predicted by general function prediction, posttranslational modification, protein turnover, chaperones, secondary metabolites biosynthesis, transport and catabolism, signal transduction mechanisms, lipid transport and metabolism, amino acid transport and metabolism. The KEGG classification of differentially expressed genes indicated that differentially expressed genes are mainly involved in carbohydrate metabolism, translation, amino acid metabolism, lipid metabolism, transport and catabolism, signal transduction, folding, sorting and degradation.
