摘 要:以贡菊试管苗叶片为外植体,研究了影响不定芽分化和生根的因素,旨在建立贡菊高效的再生体系,同时探究了羧苄青霉素浓度对叶片不定芽分化的影响,为贡菊育种奠定基础。结果表明:在MS+BA 2.0 mg/L+NAA 0.6 mg/L的培养基中,贡菊试管苗叶龄为25 d的叶片中愈伤组织诱导和不定芽分化率达到最高(88.4%),每个叶盘平均分化4.6个不定芽。当MS培养基中的IBA浓度为0.25 mg/L时,最适合贡菊试管苗根的生长,根长为4.5 cm,根数达15条,同时地上部茎叶生长也较好。低于250 mg/L的羧苄青霉素不影响不定芽愈伤组织中的诱导,但浓度高于500 mg/L时,抑制不定芽的分化。In order to establish an efficient regeneration protocol for Gongju, medium seedling leaves of Gongju as matereials, studies of elements impacting on plantlet regeneration by leaves induced calli and adventitious bud of Gongju were conducted, as well as the effects of carbenicillin concentration on adventitious bud differentiation of Gongju leaves were screened. The results showed that at 25 days of leaf age, adventitious bud differentiation and average differentiation of each leaf disc reached a maximum of 88.4% with 4.6 cultures on medium of MS+BA2.0 mg/L+NAA0.6 mg/L. When the concentration of IBA was 0.25 mg/L, it was suitable for the growth of adventitious roots of Gongju. The root length 4.5 cm and root number 15 roots were the highest. The induction rate of adventitious bud and the average number of adventitious buds in the leaf disc were not significantly different when the concentration of carbenicillin lower than 250 mg/L and without carbenicillin of medium, wheras the concentration was higher than 500 mg/L, the differentiation of adventitious buds was inhibited.
