摘  要:目的建立HPLC法同时测定不同红花Carthamus tinctorius L.(Safflower)中羟基红花黄色素A、芦丁、木犀草素、槲皮素的含有量。方法红花60%甲醇提取物的分析采用Agilent ZORBAX SB-C18色谱柱(4.6 mm×150 mm,5μm);流动相乙腈-0.5%乙酸,梯度洗脱;体积流量0.5 mL/min;柱温25℃;检测波长300 nm。结果羟基红花黄色素A、芦丁、木犀草素、槲皮素分别在17.36~282(R2=0.999 7)、1.68~181.84(R2=0.999 5)、0.52~8.71(R2=0.999 5)、5.17~94.68(R2=0.999 4)μg/mL的范围内线性关系良好,平均加样回收率96.83%~98.39%,RSD 1.13%~2.36%。结论该方法准确稳定,重复性好,可用于红花的质量控制。AIM To establish an HPLC method for the simultaneous content determination of hydroxysafflor yellow A,rutin,luteolin and quercetin in Carthamus tinotorius L.(Safflower).of different varieties.METHODS The analysis of 60%methanol extract of C.tinotorius was performed on a 25℃thermostatic Agilent ZORBAX SB-C18 column(4.6 mm×150 mm,5μm),with the mobile phase comprising of acetonitrile-0.5%acetic acid flowing at 0.4 mL/min in a gradient elution manner,and the detection wavelength was set at 300 nm.RESULTS Hydroxysafflor yellow A,rutin,luteolin and quercetin showed good linear relationships within the ranges of 17.36-282(R2=0.9997)、1.68-181.84(R2=0.9995)、0.52-8.71(R2=0.9995)、5.17-94.68(R2=0.9994)μg/mL,whose average recoveries were 96.83%-98.39%with the RSDs of 1.13%-2.36%,respectively.CONCLUSION This accurate,stable and reproducible method can be used for the quality control of C.tinotorius.
