β-catenin和cyclin D1在人泡球蚴感染病灶旁肝细胞中的表达及其意义

摘　　要：目的探讨泡球蚴感染宿主病灶旁肝细胞中β-catenin和cyclin D1的表达及其意义。方法采用免疫组化方法检测β-catenin和cyclin D1在32例人泡球蚴感染病灶旁肝细胞中和12例对照组肝组织中的表达情况;同时检测Ki-67和p53在病灶旁肝细胞和对照组的表达情况。结果β-catenin在23例(71.8%)泡球蚴感染病灶旁肝细胞胞膜和胞质呈异常表达,对照组12例肝细胞均呈细胞膜正常表达;cyclin D1在21例(65.7%)泡球蚴感染病灶旁肝细胞胞核呈阳性,对照组12例肝细胞均呈阴性。β-catenin与cyclin D1在病灶旁肝细胞表达具有显著相关性(P<0.05);Ki-67和p53在病灶旁肝细胞分别呈34.4%(11/32)和18.8%(6/32)阳性,对照组Ki-67和p53均呈阴性。结论Wnt/β-catenin通路中关键因子β-catenin和cyclin D1在病变周围肝细胞内呈异常表达,结合组织学改变,认为泡球蚴感染宿主病灶旁肝细胞呈再生状态,部分呈异型增生改变。Objective To investigate the expression and significance ofβ-catenin and cyclin D1 in cysticercosis affecting peripheral hepatocytes of host lesions.Methods Immunohistochemical methods were used to detect the expression ofβ-catenin and cyclin D1 in 32 cases of cysticercosis affecting peripheral hepatocytes of host lesions and 12 normal liver tissues used in the control group.The expression of Ki-67 and P53 was also detected.Resultsβ-catenin immunoreactivity was detected in 23 of 32(71.8%)cases of membranes and cytoplasm of liver cells near the lesions of cysticercosis infection,while negative expression was found in the control group.Cyclin D1 immunoreactivity was detected in 21 of 32(65.7%)cases of nucleus of liver cells near the lesions of cysticercosis infection,while negative expression was found in the control group.Ki-67 and P53 expression showed 34.3%(11/32)and 18.8%(6/32)in the lesion,respectively,while the control groups showed negative expression of Ki-67 and P53.Conclusionβ-catenin and cyclin D1,main factors of Wnt/β-catenin pathway,are abnormally expressed in liver cells around the lesions.Combined with histological changes,it is speculated that the liver cells near the lesions of the host lesions of pachinococcosis infection show a state of regeneration,and some of them show dysplasia.
