SEMA3B、AEG1蛋白在食管癌组织中的表达及临床意义

摘　　要：目的探讨食管癌组织中信号素3B(SEMA3B)蛋白、星型细胞上调基因1(AEG1)蛋白的表达情况及临床意义。方法收集120例食管癌患者的食管癌组织标本及对应癌旁组织标本,采用免疫组化染色法及蛋白质印迹法(Western blot)检测两种组织中SEMA3B、AEG1蛋白的表达情况,并对食管癌组织中SEMA3B、AEG1蛋白表达与食管癌患者临床特征的关系进行分析。结果食管癌组织中SEMA3B蛋白的阳性表达率和相对表达量均明显低于癌旁组织(P﹤0.01);食管癌组织中AEG1蛋白的阳性表达率和相对表达量均明显高于癌旁组织(P﹤0.01)。TNM分期为Ⅲ~Ⅳ期、有淋巴结转移食管癌患者食管癌组织中SEMA3B蛋白的阳性表达率分别低于TNM分期为Ⅰ~Ⅱ期、无淋巴结转移的患者(P﹤0.05);TNM分期为Ⅲ~Ⅳ期、分化程度为低分化、有淋巴结转移食管癌患者食管癌组织中AEG1蛋白的阳性表达率分别高于TNM分期为Ⅰ~Ⅱ期、分化程度为高中分化、无淋巴结转移的患者(P﹤0.05)。结论食管癌组织中SEMA3B蛋白表达下调,AEG1蛋白表达上调,且SEMA3B、AEG1蛋白表达与食管癌患者的病情进展有关。Objective To investigate the expression and clinical significance of semaphorin 3B(SEMA3B)protein and astrosyte elevated gene 1(AEG1)protein in esophageal cancer tissues.Method The esophageal cancer tissue specimens and surrounding tissue specimens from 120 patients with esophageal cancer were collected.The expressions of SEMA3B and AEG1 protein in these specimens were detected by immunohistochemical staining and Western blot.The relationship between the expression of SEMA3B and AEG1 protein in esophageal cancer tissues and the clinical characteristics of patients with esophageal cancer was analyzed.Result The positive expression rate and relative expression of SEMA3B protein in esophageal cancer tissues were significantly lower than those in surrounding tissues(P<0.01);the positive expression rate and relative expression of AEG1 protein in esophageal cancer tissues were significantly higher than those in surrounding tissues(P<0.01).The positive expression rate of SEMA3B protein in esophageal cancer tissues with TNM stage Ⅲ~Ⅳ and positive lymph node metastasis was lower compared to those with TNM stage Ⅰ~Ⅱ and no lymph node metastasis(P<0.05);the positive expression rate of AEG1 protein was higher in esophageal cancer tissues with TNM stage Ⅲ~Ⅳ,poor differentiation and positive lymph node metastasis compared to tissues with TNM stage Ⅰ~Ⅱ,moderate to high differentiation,and no lymph node metastasis(P<0.05).Conclusion The expression of SEMA3B protein was down-regulated while AEG1 protein was up-regulated in esophageal cancer tissues,furthermore,the expression of SEMA3B and AEG1 protein are closely related to the progression of esophageal cancer.
