摘  要:【目的】探讨多西他赛联合异环磷酰胺对卵巢癌CAOV3细胞周期蛋白D1(cyclinD1)及其生长抑制和DNA损伤诱导蛋白153(GADD153)表达的影响.【方法】以购置的卵巢癌CAOV3细胞分为三份,依次记录为空白对照组、多西他赛组及联合用药组.空白对照组常规加入培养液进行培养,多西他赛组单一给予多西他赛干预,联合用药组为多西他赛联合异环磷酰胺进行干预,三组均进行72 h干预,采用MTT法测定三组不同时间点细胞增殖率;采用PI单染流式细胞术完成两组细胞周期的测定;采用Western Blot法检测三组cyclinD1及GADD153表达水平.【结果】联合用药组培养后24 h、36 h及72 h细胞增殖率均低于多西他赛组与空白对照组(P<0.05);联合用药组卵巢癌CAOV3细胞G0/G1期低于多西他赛组和空白对照组,细胞S期高于多西他赛组和空白对照组(P<0.05);联合用药组细胞培养后24 h、36 h、72 h cyclinD1水平均低于多西他赛组和空白对照组,GADD153表达水平高于多西他赛组和空白对照组(P<0.05).【结论】多西他赛联合异环磷酰胺能诱导、促进卵巢癌CAOV3细胞凋亡,有助于改善cyclinD1及GADD153表达水平.【Objective】To investigate the effects of docetaxel combined with ifosfamide on cyclin D1,growth inhibition and DNA damage-inducible protein 153(GADD153)expression in ovarian cancer CAOV3 cells.【Methods】The ovarian cancer CAOV3 cells were divided into three groups,which were recorded as blank control group,docetaxel group and combination group.Cells in the blank control group was routinely added with regular tissue culture medium.The docetaxel group was given single docetaxel intervention and the combination group was treated with docetaxel and ifosfamide.All treatments in three groups were applied for 72 hours.The cell proliferation rates at different time points were measured by MTT assay.The cell cycle was determined by flow cytometry with PI staining.The expression levels of cyclinD1 and GADD153 were detected by Western Blot.【Results】The cell proliferation rate of the combination group was higher than that of the docetaxel group and the blank control group at 24h,36h and 72h(P<0.05).The G0/G1 phase of ovarian cancer CAOV3 cells in the combination group was lower than that in the docetaxel group and the blank control group;and the S phase in the combination group was higher than that in docetaxel group and blank control group(P<0.05).The level of cyclin D1 was lower but GADD153 was higher than those in the docetaxel group and the blank control group at 24h,36h and 72h after intervention(P<0.05).【Conclusion】Docetaxel combined with ifosfamide can induce and promote apoptosis of ovarian cancer CAOV3 cells through expression changes of cyclin D1 and GADD153 proteins.
