摘 要:目的观察红景天苷对急性胰腺炎(AP)大鼠胰腺损伤的影响,并探讨其作用机制。方法SPF级雄性Wistar大鼠144只,随机分为对照组、模型组、治疗组,每组48只。治疗组和模型组采用胆胰管逆行注射2.0%牛磺胆酸钠建立AP模型,对照组不制作AP模型。治疗组于造模后即刻经腹腔注射红景天苷4.0 mg/kg、2次/d,模型组和对照组注射等量生理盐水。分别于造模后6、12、24、48、72、96 h检测各组大鼠血清淀粉酶水平,取胰腺组织进行组织病理学评分,采用ELISA法检测胰腺组织中的Ⅰ型胶原(ColⅠ),采用荧光定量PCR法检测胰腺组织中的转化生长因子(TGF)β1、TGF-β1Ⅱ型受体(TβRⅡ)、Smad3 mRNA。结果模型组造模后各时点淀粉酶水平均高于对照组(P均<0.05)。治疗组造模后各时点淀粉酶水平均低于模型组(P均<0.05)。治疗组和模型组胰腺组织病理学评分高于对照组,治疗组低于模型组(P均<0.05)。模型组造模后各时点胰腺组织中ColⅠ含量均高于对照组(P均<0.05)。治疗组造模后12、48、72、96 h ColⅠ含量低于模型组(P均<0.05)。模型组造模后12、24、48、72、96 h胰腺组织中TGF-β1、TβRⅡ、Smad3 mRNA相对表达量均高于对照组(P均<0.05)。治疗组造模后12、48、72、96 h TGF-β1、TβRⅡ、Smad3 mRNA相对表达量低于模型组(P均<0.05)。结论红景天苷可减轻AP大鼠胰腺损伤,并促进其修复,作用机制可能与抑制TGF-β1/Smad3信号通路持续激活有关。Objective To observe the effect of salidroside on pancreatic injury in rats with acute pancreatitis(AP)and to explore its mechanism.Methods Totally 144 SPF male Wistar rats were randomly divided into the control group,model group,and treatment group.AP model was established by retrograde injection of 2.0% sodium taurocholate into biliopancreatic duct.Salidroside(4.0 mg/kg,twice a day)was injected into the abdominal cavity immediately after the model establishment in the treatment group,and normal saline was injected into the control group and the model group,respectively.Serum amylase activity was measured at 6,12,24,48,72,and 96 h after modeling.Type I collagen(ColI)content was measured by ELISA.The mRNA expression of transforming growth factorβ1(TGF-β1),TGF-β1Ⅱreceptor(TβRⅡ),and Smad3 in the pancreatic tissues was detected by fluorescence quantitative PCR.Results The amylase activity of the model group at each time point was higher than that in the control group(P<0.05).The amylase activity of the treatment group at each time point was lower than that in the model group(P<0.05).The histopathological score of pancreas in the treatment group and the model group was higher than that in the control group,and that in the treatment group was lower than that in the model group(both P<0.05).The content of ColI in the pancreas tissues of the model group was higher than that of the control group(P<0.05).The ColI level of pancreas tissues in the treatment group at 12,48,72,and 96 h after molding was lower than that of model group(all P<0.05).The relative mRNA expression of TGF-β1,TβRⅡ,and Smad3 in the model group at 12,24,48,72,96 h after molding was higher than that in the control group(all P<0.05).The relative mRNA expression of TGF-β1,TβRⅡ,and Smad3 in the treatment group was lower than that in the model group at 12,48,72,96 h after modeling(all P<0.05).Conclusion Salidroside can alleviate the pancreatic injury and promote the repair of AP,and the mechanism may be related to the inhibition of TGF-β1/Sm
