摘 要:间充质干细胞(mesenchymal stem cell, MSC)具有多向分化潜能和免疫调节能力,而且没有免疫原性,在临床上应用广泛,可以用于同种异体治疗多种自身免疫性疾病和组织修复等。但是干细胞的长距离运输和保存问题给间充质干细胞的广泛应用带来了一定的困难。目前,运输过程中时间和温度对干细胞存活率的影响不是十分明确。本实验以6组第4代骨髓间充质干细胞为例,采集细胞清洗后重悬于密封的生理盐水瓶中,可以直接用于临床注射。对注射前的细胞进行细胞表面标记和分化潜能鉴定,同时检测0℃、4℃、冰盒环境(8℃-17℃)、常温和37℃等不同的环境温度下干细胞随着时间的延长存活率的变化。实验结果显示,在设置温度为4℃的小型车载冰箱中运输的干细胞状态保存最好,在6小时内细胞存活率可以保持在90%以上,在保温箱中放置冰袋(8℃-17℃)保存的效果次之,在2小时内细胞存活率可以保持在90%以上。因此,推荐在超过2小时并保持在6小时内的长距离运输过程中,用4℃恒温小型冰箱保持干细胞的活性。如果运输时间超过6小时,不建议进行远距离运输,应该选择更快捷的交通工具,让运输过程保持在6小时内。Mesenchymal stem cells( MSCs) have multidirectional differentiation potential and immunomodulatory ability, but no immunogenicity. It is widely used in clinical, and can be used for allogeneic treatment of various autoimmune diseases and tissue repair. However, the long distance transport and preservation of stem cells have brought some difficulties to the application of mesenchymal stem cells widely. At present, the transit time and temperature on the impact of the survival rate of stem cells is not very clear. This experiment sets six groups of bone marrow mesenchymal stem cells on the fourth generation as an example. The suspended cells were acquired after cleaning with saline, and placed in physiological saline before injection, and can be directly used for clinical treatment. We identified cell surface markers and differentiation potential of MSCs before injection, meanwhile detecting the survival rate of MSCs under different ambient temperature of 0 ℃, 4 ℃, ice boxes environment(8 ℃-17℃), room temperature and 37℃ with the extension of time. The results showed that the stem cells transported in a small vehicle-mounted refrigerator at 4℃ were the best preserved, with a cell survival rate of more than 90% within 6 hours, followed by placing an ice bag(8℃-17℃) in an incubator, with a cell survival rate of more than 90% within 2 hours. Therefore, it is recommended to maintain stem cell activity in a small refrigerator with constant temperature of 4℃ during a long distance transportation between 2 hours and 6 hours. If the transportation time is more than 6 hours, it is not recommended to carry out the distance transportation, but a faster transportation should be chosen to keep the transportation process within 6 hours.
