摘  要:目的检测赖氨酸特异性去甲基化酶(LSD1)在前列腺癌组织中的表达,并探讨其对前列腺癌细胞上皮间质转化和侵袭能力的影响。方法通过免疫组化观察LSD1在高Gleason评分、低Gleason评分的前列腺癌及良性前列腺增生组织中的表达;将前列腺癌LNCaP细胞转染LSD1-过表达质粒(过表达组)或siRNA(干扰组)来上调或下调LSD1的表达,采用实时定量聚合酶链反应(real-time PCR)进行验证;用Transwell细胞迁移和侵袭试验评估LSD1表达的上调或下调对前列腺癌LNCaP细胞侵袭力的影响,并用蛋白质印迹技术检测LSD1和上皮间质转化标记物E-钙黏素(E-cadherin)、N-钙黏素(N-cadherin)和α-平滑肌动蛋白(α-SMA)的表达。结果LSD1在前列腺癌组织内呈高表达,明显高于良性前列腺增生组织,且其在高Gleason评分的前列腺癌组织中的表达显著高于低Gleason评分者与空白对照组比较,LSD1基因和蛋白表达水平在过表达组中显著增加(P<0.05),LSD1表达增加显著提高前列腺癌LNCaP细胞的迁移和侵袭能力,并且使N-cadherin和α-SMA的表达水平显著增加(P<0.05),而使E-cadherin的表达水平显著下降(P<0.05);与空白对照组比较,LSD1基因和蛋白表达水平在干扰组中显著降低(P<0.05),抑制LSD1表达显著降低前列腺癌LNCaP细胞的迁移和侵袭能力,并且使N-cadherin和α-SMA的表达水平显著减少(P<0.05),而使E-cadherin的表达水平显著增加(P<0.05)。结论LSD1在前列腺癌中表达增加,能够促进前列腺癌细胞上皮间质转化,并增加前列腺癌细胞的侵袭能力。ObjectiveTo investigate the effects of LSD1 on the epithelial-mesenchymal transition(EMT)and invasion of prostate cancer(PCa)cells.MethodsThe expressions of LSD1 in PCa with high and low Gleason scores and benign prostatic hyperplasia(BPH)were detected with immunohistochemistry.After LSD1-ORF plasmid and siRNA were transfected into LNCaP cells respectively to up-and down-regulate the expression of LSD1,the change of LSD1 expression was determined with Real-time PCR,the migration ability of LNCaP cells was detected with Transwell assay,and the expressions of LSD1,E-cadherin,N-cadherin andα-SMA were determined with Western blot.ResultsPCa specimens had higher LSD1 expression than BPH specimens,and LSD1 expression was positively correlated with Gleason score.Compared with the control group,LSD1-ORF group had significantly up-regulated LSD1 expression(P<0.05).Up-regulated LSD1 increased the migration and invasion ability of LNCaP cells and the expressions of N-cadherin andα-SMA(P<0.05),but decreased the expression of E-cadherin(P<0.05).Compared with the control group,the siRNA group had significantly down-regulated LSD1 expression(P<0.05).Down-regulated LSD1 decreased the migration and invasion ability of LNCaP cells and the expressions of N-cadherin andα-SMA(P<0.05),but increased the expression of E-cadherin(P<0.05).ConclusionLSD1 is highly expressed in PCa tissues,which can promot the EMT process and increase the invasion of prostate cancer cells.
