摘 要:目的探讨肿瘤坏死因子(TNF-α)对人前列腺癌ARCaP细胞上皮细胞间质转化的影响。方法比较人前列腺癌ARCaPE细胞和ARCaPM细胞形态学差异,通过Western blot实验比较ARCaPE细胞和ARCaPM细胞E-cadherin表达水平差异;通过Transwell体外侵袭实验比较ARCaPE细胞和ARCaPM细胞体外侵袭能力差异;通过real time RT-PCR实验比较ARCaPE细胞和ARCaPM细胞TNF-α表达水平差异;通过重组人TNF-α蛋白处理ARCaPE细胞,观察比较处理组与对照组在形态学、E-cadherin表达及体外侵袭能力方面的差异。结果ARCaPE细胞呈类似铺路石样的类圆形,细胞间连接紧密,呈岛屿状生长。ARCaPM细胞排列松散,部分细胞拉长呈纺锤形。与ARCaPE细胞相比,ARCaPM细胞E-cadherin表达下调,但具有更高的体外侵袭能力和更高的TNF-α表达水平。通过重组人TNF-α蛋白处理ARCaPE细胞,可使其排列松散,细胞拉长呈纺锤形,E-cadherin表达下调,并获得较高体外侵袭能力。结论TNF-α可诱导人前列腺癌ARCaP细胞EMT。ObjectiveTo investigate the effects of tumor necrosis factorα(TNF-α)on epithelial-mesenchymal transition(EMT)in human prostate cancer ARCaP cells.MethodsMorphology of human prostate cancer ARCaPE cells and ARCaPM cells were compared.E-cadherin expression and invasive capacity of ARCaPE and ARCaPM cells were determined with Western blot and Transwell assay,respectively.The expression of TNF-αin ARCaPE cells and ARCaPM cells were compared with real time RT-PCR.ARCaPE cells were treated with recombinant human TNF-α,and the differences in morphology,E-cadherin expression and invasive capacity between the treated and control cells were observed.ResultsARCaPE cells were cobblestone-like,closely connected and growing like islands,while ARCaPM cells were spindle-shaped and loosely connected.Compared with ARCaPE cells,ARCaPM cells had decreased E-cadherin expression,higher invasive capacity,and higher TNF-αexpression.TNF-αtreatment changed the morphology of ARCaPE cells from cobblestone-like to spindle-shaped,made them loosely connected,downregulated E-cadherin expression,and enhanced the invasive capacity.ConclusionTNF-αcan induce EMT in human prostate cancer ARCaP cells.
