汉防己碱对人膀胱癌细胞增殖、凋亡和侵袭的影响

摘　　要：目的分析汉防己碱(Tet)对人膀胱癌细胞增殖、凋亡和侵袭的影响,并初步探讨其分子机制。方法利用结晶紫染色和克隆形成实验检测细胞增殖;Transwell实验检测细胞侵袭;Hochest 33258染色和Western blot检测细胞凋亡;Western blot检测Wnt/β-catenin信号的活化情况。结果汉防己碱可抑制人膀胱癌细胞增殖和侵袭,并促进细胞凋亡。汉防己碱可抑制DVL蛋白表达,抑制GSK3β在9位丝氨酸的磷酸化,并降低β-catenin及其下游靶分子Cyclin D1和c-Myc的蛋白水平。结论汉防己碱可抑制人膀胱癌细胞增殖和侵袭,并促进细胞凋亡,这可能与其对Wnt/β-catenin信号的抑制作用有关。Objective To determine the effect of tetrandrine on the proliferation,invasion and apoptosis of human bladder cancer cells,and the possible mechanism involved.Methods Cell proliferation was determined by crystal violet staining and colony formation assay.Cell apoptosis was detected by Hochest 33258 staining and Western blot.Cell invasion was analyzed by transwell assay.The activation of Wnt/β-catenin signal was analyzed by Western blot.Results Tetrandrine inhibited the proliferation and invasion of human bladder cancer cells,and promoted the cell apoptosis.It also decreased DVL protein expression,suppressed phosphorylation of GSK3βat serine 9(Ser 9),and reduced the protein levels ofβ-catenin and its downstream target molecules Cyclin D1 and c-Myc.Conclusion Tetrandrine may inhibit the proliferation,and invasion of human bladder cancer cells,and promote the cell apoptosis possibly through the inhibition of Wnt/β-catenin signals.
