摘 要:目的探讨肝癌高表达(HULC)长链非编码RNA在肝癌中的表达及其与自噬的关系。方法采用实时定量PCR检测60例肝癌组织以及相应正常肝组织中HULC的表达水平,并检测HULC在肝癌细胞系及正常肝细胞系中的表达情况。通过pcDNA3.1-HULC转染肝癌细胞系HepG2、Bel-7402过表达HULC;分别采用CCK-8试剂盒、Western blotting和免疫荧光检测细胞增殖改变情况、自噬相关蛋白LC3Ⅱ/Ⅰ和自噬相关蛋白Atg3的表达变化以及LC3斑点数目的变化情况。结果 HULC在肝癌组织中的表达高于正常肝组织,差异有统计学意义(P <0.05)。过表达HULC能促进肝癌细胞增殖,并且过表达HULC的肝癌细胞中LC3-Ⅰ向LC3-Ⅱ的转化和Atg3的表达均显著增加(P <0.05)。免疫荧光可见,过表达HULC后LC3荧光斑点明显增多。结论肝癌组织中HULC的表达高于正常肝组织,HULC促进肝癌细胞增殖,并且能够提高肝癌细胞自噬水平,有望成为肝癌治疗的潜在靶点。Objective To investigate the expression of the long non-coding RNA(lncRNA) HULC and its influence on autophagy in hepatocellular carcinoma(HCC). Methods Real-time PCR was used to detect HULC expression in 60 HCC samples,adjacent normal tissues,and HCC and normal cell lines. HepG2 and Bel-7402 cells were transfected with pcDNA3.1-HULC,and CCK-8 assays,Western blotting,and immunofluorescence were used to detect cell proliferation changes,expressions of LC3Ⅱ/Ⅰ and Atg3 protein,and number of LC3 fluorscence points,respectively. Results Compared with normal tissues,HULC expression was upregulated in HCC tissues(P < 0.05). Overexpressing HULC in HCC cells significantly upregulated the expression of LC3-Ⅱ to LC3-Ⅰ and autophagy-related protein Atg3(P < 0.05). Immunofluorescence assays showed that HULC expression can increase the number of LC3 fluorescence points. Conclusion HULC expression is higher in liver cancer tissues than in normal tissues. HULC may promote proliferation of,and autophagy within HCC cells,and thus may be a viable target for treatment of HCC.
