摘 要:目的测定不同产地及不同采收期青钱柳叶蛋白含量,探讨其体外降血糖和抗氧化活性。方法采用考马斯亮蓝G-250溶液染色法测定青钱柳叶中蛋白含量;采用α-葡萄糖苷酶抑制法研究该蛋白体外降血糖作用;以抗坏血酸(VitC)为阳性对照,采用1,1-二苯基-2-三硝基苯肼自由基(DPPH·)清除法、2,2’-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基清除法和铁离子还原/抗氧化能力(FRAP)法评价其体外抗氧化活性。结果不同产地25批药材中蛋白含量范围为0.58%~1.39%,同一产地不同采收期药材蛋白含量差异较小。青钱柳叶蛋白对α-葡萄糖苷酶具有抑制作用,IC50为0.0158 g·L^-1;对DPPH、ABTS自由基具有清除能力,IC50分别为0.1368、0.0696 g·L^-1,对Fe3+具有还原能力,FRAP为9.98×10^3μmol·g^-1。青钱柳叶蛋白对ABTS自由基的清除能力和对Fe3+还原能力均比VitC强。结论不同产地青钱柳叶蛋白的含量有差异,该蛋白具有明显的体外降血糖和抗氧化活性。Objective To compare the proteins’contents in the leaves of Cyclocarya paliurus from different habitats and different harvesting periods.Methods The protein content in the leaves of Cyclocarya paliurus was determined by Coomassie brilliant blue G-250 solution staining method.The hypoglycemic activity of the protein in vitro was studied by alpha-glucosidase inhibition method.Using VitC as positive control,the DPPH·scavenging method,ABTS free radical scavenging method,and FRAP method were applied to evaluate the in vitro antioxidant activity.Results The protein contents ranged from 0.58%to 1.39%in 25 batches of medicinal materials from different origins.Little difference was found in protein contents between different harvesting periods of medicinal materials from the same origin.Protein of Cyclocarya paliurus leaves inhibited alpha-glucosidase with an IC50 of 0.0158 g·L^-1;and scavenged DPPH and ABTS free radicals,with IC50 of 0.1368 and 0.0696 g·L^-1,respectively.It also reduced Fe3+in FRAP at 9.98×10^3μmol·g^-1.ABTS radical scavenging capacity and Fe3+reduction capacity of protein of Cyclocarya paliurus leaves were stronger than VitC.Conclusion The contents of proteins of Cyclocarya paliurus leaves from different habitats were varied.The proteins had obvious hypoglycemic and antioxidant activities in vitro.
