治糜康栓对宫颈炎模型大鼠IL-1β、EGFR及TNF-α表达的影响

摘　　要：目的探讨治糜康栓治疗宫颈炎大鼠的疗效与机制。方法用苯酚胶浆阴道注射建立宫颈炎大鼠模型,60只大鼠随机分为空白组、模型组、基质组、阳性组(市售治糜康栓)和改良工艺后治糜康栓低、高剂量组。治疗12天以后,用试剂盒检测大鼠血清中表皮生长因子受体(EGFR)、白细胞介素1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的含量。HE染色观察大鼠宫颈病理组织变化,免疫组织化学检测宫颈组织中TNF-α、EGFR和IL-1β含量。结果与空白组比较,模型组TNF-α、IL-1β含量增多(P<0.01),EGFR含量减少(P<0.01)。与模型组比较,高剂量组TNF-α、IL-1β含量减少(P<0.01),EGFR含量增多(P<0.01)。结论治糜康栓下调TNF-α、IL-1β介导的炎症反应,上调EGFR的表达,调节宫颈炎大鼠的免疫功能,这可能是其治疗宫颈炎的机制之一。Objective To investigate the efficacy and mechanism of Zhimikangshuan in the treatment of cervicitis rats. Methods A rat model of cervicitis was established by vaginal injection of phenol colloid. Sixty rats were randomly divided into blank group, model group, stromal group, positive group(commercially-resolved of Zhimikangshuan) and modified technique to treat low-and high-dose groups of of Zhimikangshuan suppository. After 12 days of treatment, the serum levels of epidermal growth factor receptor(EGFR), interleukin-1β(IL-1β), and tumor necrosis factor-α(TNF-α) were measured using a kit. The pathological changes of cervical tissues were observed by HE staining. The contents of TNF-α, EGFR and IL-1β in cervical tissues were detected by immunohistochemistry.Results Compared with the blank group, the TNF-α and IL-1β levels in the model group increased(P<0.01) and the EGFR content decreased(P<0.01). Compared with the model group, the levels of TNF-α and IL-1β were decreased in the high-dose group(P<0.01), and the EGFR content was increased(P<0.01). Conclusion Zhimikangshuan suppository can down-regulate TNF-α and IL-1β-mediated inflammation, up-regulate the expression of EGFR, and regulate the immune function of cervicitis rats, which may be one of the mechanisms for the treatment of cervicitis.
