摘 要:目的研究CK34βE12、Galectin-3在甲状腺乳头状癌(PTC)中的表达以及与BRAFV600E基因突变的关系,评价其在甲状腺乳头状癌中的临床诊断价值。方法收集2017年6月~2018年6月在我院接受治疗的230例甲状腺乳头状癌患者的甲状腺乳头癌组织及其癌旁组织,采用免疫组织化学法检测CK34βE12、Galectin-3的表达情况,AMERS法检测BRAFV600E,采用χ2检验方法分析CK34βE12、Galectin-3蛋白阳性率与BRAFV600E基因突变率关系。结果在PTC组织中,CK34βE12、Galectin-3呈强阳性表达(P<0.01),与患者年龄、性别、是否淋巴结转移等均无关联(P>0.05);BRAFV600E基因在PTC中有84.3%突变;BRAFV600E基因突变率与患者年龄、性别、是否淋巴结转移等无关联(P>0.05),与CK34βE12、Galectin-3蛋白表达无关联(P>0.05)。结论 PTC组织中,CK34βE12、Galectin-3对鉴别甲状腺良恶性具有重要临床意义,但CK34βE12、Galectin-3表达升高与BRAFV600E基因突变的相互关系,还需要进一步研究。Objective To investigate the expression of CK34βE12 and Galectin-3 in papillary thyroid carcinoma(PTC) and its relationship with BRAFV600E gene mutation, so as to evaluate its clinical diagnostic value in papillary thyroid carcinoma. Methods 230 cases of thyroid papillary carcinoma and adjacent tissues who were treated in our hospital from June 2017 to June 2018 were collected. The expression of CK34βE12 and Galectin-3 was detected by immunohistochemistry. The BRAFV600E gene was detected by the AMERS method. The χ2 test method was used to analyze the relationship between the positive rate of CK34βE12 and Galectin-3 protein and the mutation rate of BRAFV600E gene.Results CK34βE12 and Galectin-3 were strongly positively expressed in PTC tissues(P<0.01), and were not associated with age, sex, and lymph node metastasis(P>0.05), the BRAFV600E gene showed 84.3% mutations in PTC;the BRAFV600E gene mutation rate was not associated with age, gender, lymph node metastasis(P>0.05), and was not associated with CK34βE12 and Galectin-3 protein expression(P>0.05). Conclusion CK34βE12 and Galectin-3 in PTC tissues have important clinical significance in the differential diagnosis of benign and malignant thyroid. However, the relationship between the increased expression of CK34βE12 and Galectin-3 and the mutation of BRAFV600E gene needs further study.
