摘 要:微生物固态发酵法是简易有效的饲料中单宁处理方法之一。筛选微生物资源是微生物固态发酵法的核心内容。自青海门源高寒草甸土壤中分离到一株单宁降解菌DT-3,对菌种DT-3进行了鉴定,并针对青海“双低”菜籽粕完成固态发酵处理效果研究。结果显示,菌种DT-3具有明显的单宁降解特性。通过形态学检测、生理生化检测及16S rRNA序列比对分析,菌种DT-3属于假黄单胞菌属(Pseudoxanthomonas),命名为Pseudoxanthomonas sp.DT-3(MK 418539)。通过DT-3对青海“双低”菜籽粕中单宁降解效果的研究表明,在料液比、发酵温度及发酵天数等处理条件下,DT-3单宁降解效果差异显著,其中料液比100%、发酵温度25℃、发酵天数5 d时单宁降解率最大,分别达到58.8%、62.95%及61.74%。综上所述,菌种DT-3是有效的“双低”菜籽粕固态发酵处理备选菌种资源。Solid-state fermentation based on microbial resources is one of most effective methods for tannin treatment.Screening degrading microbial resource is an important part of fermentation.A tannin-degrading strain DT-3 was isolated from alpine meadow soil,Men Yuan,Qinghai.DT-3 was identified in this study to exploit the potential of the strain for tannin treatment.The degrading properties to Qinghai“double-low”rapeseed meal were researched as well.In this study,strains inhabited in Qinghai typical ecosystems were isolated by pour plate method and streak plate method,and the tannin-degrading strain was screened by agar selecting method.The morphology,biochemical characteristic and 16S rRNA sequence were analyzed for identification the purpose strain.For evaluating the strain’s capability of tannin degradation,solid-liquid ratio,temperature and time as the main influencing factors were chose to analyze by spectrophotometer method.As a result,123 strains were isolated from alpine meadow soil,Men Yuan,Qinghai.A tannin-degrading strain named DT-3 was researched.According to the morphological detection,physiological and biochemical detection and 16S rRNA sequence alignment analysis were performed,DT-3 belongs to Pseudoxanthomonas,and was named Pseudoxanthomonas sp.DT-3(MK 418539).In addition,DT-3’s capability for catalyzing tannin in Qinghai“double-low”rapeseed meal was evaluated as follows:under different conditions,DT-3’s tannin degradation effect showed significant differences.100%solid-liquid ratio,25℃and 5 d were optimum treatment conditions,in which the degradation rate of tannin reached 58.8%,62.95%and 61.74%respectively.To sum up,DT-3 showed potential capability for biodegradation.
