摘 要:为研究miRNA表达量与肉质风味的相关性,试验采用荧光定量PCR技术(Q-PCR)检测了4个猪种背最长肌中miR-23a、miR-151、miR-299、miR-199a和miR-497的差异表达。结果表明,miR-23a、miR-151在藏猪背最长肌中表达最高,miR-299、miR-199a和miR-497在约克夏猪背最长肌中表达最高;相关性分析显示,miR-23a、mi R-151在背最长肌中的表达水平与肌内脂肪含量和硫胺素含量呈正相关,与肌纤维面积、眼肌面积和瘦肉率呈负相关,miR-299、miR-199a和miR-497在背最长肌中的表达水平与肌内脂肪含量和硫胺素含量呈负相关,与肌纤维面积、眼肌面积和瘦肉率呈正相关。综上表明,miR-23a、miR-151对猪肉品质具有正调控作用,miR-299、miR-199a和miR-497对猪胴体品质具有正调控作用。In order to investigate the correlation of miRNA expression with meat flavor phenotype index, the expression levels of miR-23 a, miR-151, miR-299, miR-199 a and miR-497 were measured in longissimus dorsi muscle of 4 pig breeds with quantitative real-time polymerase chain reactions(Q-PCR). The results showed that the expression levels of miR-23 a, miR-151 is highest in longissimus dorsi of Tibetan pigs, the expression levels of miR-299, miR-199 a and miR-497 is highest in longissimus dorsi of Yorkshire pigs;correlation analysis showed that the expression levels of miR-23 a, miR-151 were in positive relation with intramuscular fat contents and thiamine, while were negative relation with myofilament area, eye muscle area and lean meat rate. The expression levels of miR-299, miR-199 a and miR-497 were in negative relation with intramuscular fat contents and thiamine, while were positive relation with myofilament area, eye muscle area and lean meat rate. These results indicated that miR-23 a, miR-151 had positive regulation effect on pork quality, and miR-299, miR-199 a and miR-497 had positive regulation effect on pig carcass quality.
