摘 要:目的探讨人类白细胞抗原-G(HLA-G)在卵巢癌中的表达及对卵巢癌SKOV-3细胞生物学行为的影响。方法选取70例卵巢癌患者的上皮性卵巢癌组织,另选取40例正常卵巢组织,采用免疫组织化学法检测上皮性卵巢癌组织和正常卵巢组织中HLA-G表达,并对HLA-G表达与卵巢癌患者临床特征的关系进行分析。应用LipofectamineTM3000将pcDNA3.1 HLA-G真核表达载体(过表达组)及空白质粒pcDNA3.1(对照组)转染到卵巢癌SKOV-3细胞中,同时设立空白组,MTT法检测细胞活力,Transwell法检测细胞迁移及侵袭能力。结果上皮性卵巢癌组织HLA-G阳性表达率明显高于正常卵巢组织(P﹤0.01),低分化、Ⅲ~Ⅳ期和有淋巴结转移的卵巢癌患者的HLA-G阳性表达率明显高于中高分化、Ⅰ~Ⅱ期和无淋巴结转移的患者(P﹤0.01)。过表达组细胞中HLA-G蛋白相对表达量明显高于对照组和空白组细胞(P﹤0.01),细胞活力、细胞迁移及侵袭数目均明显高于对照组和空白组细胞(P﹤0.01)。结论 HLA-G在卵巢癌中过表达,并促进卵巢癌细胞的增殖、迁移及侵袭。Objective To explore the expression of human leukocyte antigen G(HLA-G) in ovarian cancer and its effect on the biological behavior of ovarian SKOV-3 cancer cells. Method The expression of HLA-G in the epithelial ovarian cancer tissues from 70 cases of ovarian cancer and in 40 cases of normal ovarian were detected by immunohistochemical assay. The relationship between HLA-G and the clinical characteristics of ovarian cancer was analyzed. The pcDNA3.1 HLA-G eukaryotic expression vector(overexpression group) and pcDNA3.1 blank plasmid(control group) were transfected into SKOV-3 cell by LipofectamineTM3000, meanwhile a blank group was set up. Cell viability was measured by MTT assay. Cell migration and invasion ability was detected by Transwell assay. Result Compared with normal ovarian tissues, the expression of HLA-G in ovarian cancer was significantly up-regulated(P<0.01). The positive expression rate of HLA-G in patients with lower degree of differentiation, stage III-IV disease and positive lymph node metastasis was significantly higher compared to those with moderate to high degree of differentiation, stage I^II disease and negative lymph node metastasis(P<0.01), and the positive expression rate of HLA-G in overexpression group was significantly higher than that in control group and blank group(P<0.01). Additionally, the cell viability, cell migration and invasion ability were evidently higher in overexpression group than in the other two groups(P<0.01). Conclusion HLA-G is over-expressed in ovarian cancer cells, and may promote the proliferation, migration and invasion of cancer cells.
