摘  要:柞蚕饰腹寄蝇Blepharipa tibialis是柞蚕主要寄生性害虫。扩增得到柞蚕饰腹寄蝇18个种群、长度为554 bp的线粒体Cytb基因片段,生物信息学分析显示其碱基A、T、C和G的平均含量分别为36.10%、42.60%、12.82%和8.48%,AT含量达78.7%,存在明显的AT使用偏向性。样本两两间遗传距离为0~0.000 30,平均遗传距离为0.000 25。柞蚕饰腹寄蝇98个个体中存在6个单倍型,单倍型多样度(H_d)为0.160,平均核苷酸差异数(K)为0.165,个体之间差异性较小。基于Cytb序列片段构建的NJ进化树显示,柞蚕饰腹寄蝇等32种双翅目昆虫从科的水平上与传统分类系统一致,按与柞蚕饰腹寄蝇的亲缘关系由近及远依次将其归类到寄蝇科、蝇科、花蝇科、麻蝇科和丽蝇科。结果表明,柞蚕饰腹寄蝇种群进化时间较短,但线粒体Cytb基因仍能用于双翅目昆虫科级进化的辅助分析。Blepharipa tibialis is the major parasitic pest of Chinese oak silkworm. In present study,mitochondrial Cytb gene fragment with a length of 554 bp was cloned from 18 populations of B. tibialis. Bioinformatical analysis showed that average content of A,T,C and G was 36. 10%,42. 60%,12. 82% and 8. 48%,respectively,presenting significant bias in AT content( 78. 7%). The pairwise genetic distances between 2 individuals ranged from 0 to 0. 000 30,and the average was 0. 000 25. There were 6 haplotype sites in 98 individuals of B. tibialis. Haplotype diversity( Hd) was 0. 160 and average nucleotide difference( K) was 0. 165,indicating only small differences could be found between individuals. NJ tree based on Cytb gene fragment showed that 32 species of Dipera insects like B. tibialis had consistent evolutionary relationship with the traditional taxonomy at family level. They were clustered into family Tachinidae,Muscidae,Anthomyiidae,Sarcophagidae and Calliphoridae in descending order according to their close relationship with B. tibialis. The study suggests that although the evolutionary process for B. tibialis is not long enough for population differentiation,mitochondrial Cytb gene can still be used for auxiliary phylogeny analysis at family level for Dipera insects.
