摘 要:目的检测原发性开角型青光眼(POAG)小梁网组织与对照眼小梁组织中自噬相关蛋白P62与LC3-Ⅱ的表达,研究自噬过程在POAG发病机制中的作用。方法对2019年1月~2019年4月在潍坊医学院附属医院眼科中心行小梁切除术的原发性开角型青光眼患者18例(20眼)进行回顾性分析。HE染色观察对照组和POAG组小梁组织的结构。免疫组化法检测对照组及POAG组小梁组织的自噬相关蛋白P62与LC3-Ⅱ的表达水平。结果 HE染色观察,POAG组小梁组织较对照组网眼变小甚至消失,小梁组织结构欠清晰,Schlemm管腔不规则性变窄,可见色素沉着。免疫组化结果显示LC3-Ⅱ在对照组小梁细胞中少量表达,在POAG组小梁细胞中表达明显增加。而P62在POAG组小梁细胞中少量表达,在对照组中表达明显增加,差异均具有统计学意义(P<0.05)。结论 POAG患者小梁网组织中P62表达水平低,LC3-Ⅱ表达水平高,自噬活性明显增加,自噬活动可能参与了POAG小梁细胞的氧化损伤。Objective To study the expressions of autophagy-related proteins P62 and LC3-Ⅱ in normal trabecular meshwork and primary open angle glaucoma(POAG) trabecular meshwork,and to explore the role of autophagy in the pathogenesis of POAG.Methods Eighteen patients(20 eyes) with POAG who underwent trabeculectomy at the department of ophthalmology of affiliated hospital of weifang medical college from January 2019 to April 2019 were retrospectively analyzed.HE staining was used to observe the structure of trabecular tissue in the control group and the POAG group and immunohistochemical method was used to detect the expression levels of autophagy-related proteins P62 and LC3-Ⅱ in trabecular tissues.Results Compare with the control group,HE showed that the trabecular tissue of the POAG group became smaller or even disappeared,the trabecular tissue structure was not clear,schlemm lumen irregularities were narrowed and pigmentation was visible.Immunohistochemistry results showed that LC3-Ⅱ was slightly expressed in trabecular meshwork in control group and increased significantly in POAG group.P62 was expressed in trabecular meshwork in POAG group and increased significantly in control group(P<0.05).Conclusion Low expression level of P62 and high level of LC3-Ⅱ in trabecular meshwork of patients with POAG,also,the activity of autophagy was significantly increase,so autophagy activity may be involved in the oxidative damage of POAG trabecular cells.
