基于TCGA数据库的RelB与前列腺癌临床相关性分析及功能验证

摘　　要：【目的】本研究拟通过癌症基因组图谱(the cancer genome atlas,TCGA)公共数据库研究RelB与前列腺癌的关联,验证其诱导前列腺癌放疗抵抗的潜在功能。【方法】在TCGA公共数据库下载前列腺癌患者RelB高通量测序数据及其对应的临床资料,运用SPSS分析RelB在前列腺癌患者中的表达水平以及临床特征,随后建立RelB沉默前列腺癌细胞株PC3,4 Gy X线进行处理,CellTiter-Glo发光法检测细胞活力。【结果】统计分析结果显示前列腺癌肿瘤组织内RelB表达显著高于前列腺增生组织,且与患者年龄、肿瘤T分期有显著性关联,虽然RelB差异表达与患者总生存无显著性关联,但在放疗人群中RelB高表达患者无复发生存期(relapse-free survival,RFS)显著低于RelB低表达组,体外实验进一步发现前列腺癌细胞株中RelB蛋白表达与细胞放疗敏感性呈显著负相关。【结论】RelB可能参与前列腺癌发生发展,且有可能是诱导前列腺癌放疗抵抗的关键因子。【Objective】To study the expression of Rel B in patients with prostate cancer and its clinical significance.【Methods】The cancer genome atlas(TCGA)public database was used to download the high-throughput sequencing data of RelB in patients with prostate cancer and its corresponding clinical data.SPSS was used to analyze the expression level and clinical characteristics of Rel B in patients with prostate cancer.Then,RelB was silenced in prostate cancer cell line PC3 and 4 Gy X-ray treatment was established for processing.Cell Titer-Glo luminescence method was used to detect the cellular viability.【Results】The statistical results showed that the expression of Rel B in prostate cancer tissues was significantly higher than that in benign prostatic hyperplasia tissues,and was significantly correlated with patient’s age and tumor T stage.Although the expression of RelB was not significantly correlated with overall survival,the relapse-free survival(RFS)in patients with high expression of Rel B was significantly lower than that in the low expression group of Rel B in radiotherapy population.Furtherly,the in vitro experiments confirmed that the expression of RelB protein in prostate cancer cell lines was negatively correlated with cell radiosensitivity.【Conclusion】Rel B may be involved in the occurrence and development of prostate cancer,and may be a key factor in inducing radioresistance of prostate cancer,which deserves further study.
