摘  要:采用超声辅助皂化法提取禾谷镰孢菌中的麦角甾醇,通过单因素和响应面分析,对影响麦角甾醇提取效果的主要因素包括皂化碱含量、提取温度、液固比及超声时间进行优化,并建立其HPLC测定方法。结果表明,禾谷镰孢菌中麦角甾醇超声提取最佳条件为碱含量0.034 g/mL、温度36℃、液固比200 mL/g、超声时间8 min,禾谷镰孢菌中麦角甾醇提取量为0.88 mg/g,与模型预测基本一致。麦角甾醇的线性范围为3.125~200μg/mL,相关系数0.9997,平均回收率在82%以上。Ergosterol in Fusarium graminearum(F.graminearum)was extracted by ultrasonic-assisted saponification.The main factors affecting the extraction efficiency of ergosterol were optimized by single factor and response surface analysis,including the content of saponification alkali,extraction temperature,liquid-solid ratio and ultrasonic time.A method for determination of ergosterol by HPLC was established.Results showed that the optimum extraction conditions of ergosterol were alkali content of 0.034 g/mL,temperature of 36℃,liquid-solid ratio of 200 mL/g,ultrasonic extraction time of8 min,and ergosterol extraction amount of 0.88 mg/g,consistent with the model prediction.The calibration curve was linear in the range of 3.125-200μg/mL with the correlation coefficients over0.9997.The average recoveries were more than 82%.
