不同针刺时长对脑出血大鼠血脑屏障保护机制的研究

摘　　要：目的:探讨不同针刺时长对脑出血(ICH)大鼠脑含水量、脑组织超微结构和水通道蛋白4(AQP-4)的影响。方法:36只雄性SD大鼠随机分为假手术组、模型组、针刺5s组、针刺60s组,检测脑组织脑含水量、电镜下脑血肿周围区超微结构、免疫组化AQP-4含量及AQP-4蛋白表达。结果:与假手术比较,模型组大脑含水量、AQP-4阳性细胞及AQP-4蛋白表达均显著上升(P<0.05);与模型组比较,针刺5s组、针刺60s组含水量下降(P<0.05),针刺60s组AQP-4阳性细胞表达显著降低(P<0.05);针刺5s组、针刺60s组AQP-4蛋白表达水平均显著降低(P<0.05)。结论:不同针刺时长均能减轻ICH后脑水肿,可降低脑组织中AQP-4蛋白表达,以60s最佳并能有效降低脑组织中AQP-4含量。Objective:To investigate the effects of different acupuncture durations on cerebral water content,the ultrastructure of cerebral tissue and aquaporin 4(AQP-4)in cerebral hemorrhage model rats.Method:Thirty-six male SD rats were randomly divided into sham group,model group,5 s acupuncture group,60 s acupuncture group,corresponding to detect cerebral water content,the ultrastructure of cerebral hematoma surrounding area,immunohistochemical AQP-4 content and AQP-4 protein expression level.Results:Compared with sham group,the water content in the brain AQP-4 positive cells and AQP-4 protein of the model group increased(P<0.05).Compared with the model group,the water content in the acupuncture 5 s group and the acupuncture 60 s group decreased(P<0.05),the expression of AQP-4 positive cells in the 60 s acupuncture group decreased significantly(P<0.05),and the expression levels of AQP-4 protein in the 5 s group and the 60 s group decreased(P<0.05).Conclusion:Different acupuncture duration can reduce cerebral edema after intracerebral hemorrhage and reduce the expression of AQP-4 protein in brain tissue,with 60 s being the better.Sixty seconds acupuncture group can effectively reduce the content of AQP-4 in brain tissue.
