摘 要:目的:观察2-羟基萨氯酚(2OH-saclofen)对慢性应激大鼠杏仁核神经元超微结构的保护作用。方法:80只雄性Wistar大鼠随机分为对照组(control)、单纯应激组(stress)、应激联合2OH-saclofen处理组(stress+2OH-saclofen)、应激联合baclofen处理组(stress+baclofen)。采用足底电击结合噪声的方法制备应激动物模型,同时分别经腹腔注射2OH-saclofen(100μg/kg)或氯苯氨丁酸(40 mg/kg)。应激40 d,灌流固定,分离出杏仁核。用透射电镜观察杏仁核内神经元超微结构的变化;高效液相-电化学法检测大鼠杏仁核内儿茶酚胺含量的变化。结果:透射电镜观察结果显示:与正常对照组比较,单纯应激组和应激联合baclofen处理组大鼠杏仁核神经元超微结构都有一定的变性改变,而应激联合2OH-saclofen处理组大鼠杏仁核神经元无明显改变。高效液相-电化学法检测显示:单纯应激组和应激联合baclofen处理组杏仁核内儿茶酚胺含量呈升高趋势,而应激联合2OH-saclofen处理组儿茶酚胺含量呈降低趋势。结论:GABAb受体抑制剂2OH-saclofen对慢性应激大鼠杏仁核神经元有保护作用。Objective: To observe the protective effect of 2OH-saclofen on ultrastructural of amygdala neurons in chronic stress rats. Methods: Eighty male Wistar rats were randomly divided into control group,simple stress group,stress + 2OH-saclofen group and stress + baclofen group. The stress animal model was prepared by plantar electric shock combined with noise. At the same time,2OH-saclofen(100 μg/kg) or baclofen(40 mg/kg) were injected intraperitoneally respectively. After 40 days of stress,the amygdala was isolated by perfusion fixation. The ultrastructure of neurons in the amygdala was observed by transmission electron microscopy. Determination of catecholamine content in amygdala of rats by high performance liquid chromatography-electrochemical method. Results: The results of transmission electron microscopy showed that compared with the normal control group,the ultrastructure of amygdala neurons in the stress group and the stress + baclofen group had certain degeneration changes. However,the amygdala neurons in stress + 2OH-sacclofen group did not change significantly. High performance liquid-electrochemical detection showed that the content of catecholamine in the amygdala of the stress group and the stress + baclofen group increased. The content of catecholamine in the stress + 2OH-saclofen group showed a decreasing trend. Conclusion: GABAb receptor inhibitor 2 OH-saclofen has a protective effect on amygdala neurons in chronic stress rats.
