摘 要:目的观察干扰ZEB1表达对结直肠癌细胞上皮间质转化与增殖、凋亡与迁移的影响。方法RT?PCR与Western Blot检测检测人结直肠癌细胞HT29、LOVO、SW480中的ZEB1的基因与蛋白表达。将对数生长期的LOVO细胞分2组转染:干扰组转染慢病毒pPLK?ZEB1?shRNA,干扰对照组转染慢病毒pPLK?control?shRNA,以未转染的细胞为正常对照。转染48 h后,检测细胞增殖、克隆形成率、迁移能力、凋亡及上皮间质转化相关蛋白表达。结果干扰ZEB1表达后,人结直肠癌细胞LOVO的增殖、克隆形成率与迁移能力明显下降(P<0.05),细胞凋亡率升高(P<0.05),上皮间质转化E?cadherin蛋白表达提高,Vimentin、Twist蛋白表达降低(P<0.05)。结论干扰ZEB1表达可抑制人结直肠癌细胞的增殖、迁移能力,增加细胞凋亡,抑制其上皮间质转化。Objective To observe the effects of ZEB1 interference on epithelial?mesenchymal transforma?tion,proliferation,apoptosis and migration of colorectal cancer cells.Methods Detection of ZEB1 gene and protein expression in human colorectal cancer cells HT29,LOVO and SW480 by RT?PCR and Western blot.Two groups of logarithmic LOVO cells were transfected:the interfering group transfected lentivirus pPLK?ZEB1?shRNA,the interfering control group transfected lentivirus pPLK?control?shRNA,and the untransfected cells as normal control.After 48 hours of transfection,cell proliferation,clone formation rate,migration ability,apoptosis and expression of epithelial?mesenchymal transition?related proteins were detected.Results After interference with ZEB1 expression,the proliferation,clone formation rate and migration ability of human colorectal cancer cell beads LOVO decreased significantly(P<0.05),the apoptotic rate increased(P<0.05),the expres?sion of E?cadherin in epithelial?mesenchymal transformation increased,and the expression of Vimentin and Twist decreased(P<0.05).Conlusion Interference of ZEB1 expression can inhibit the proliferation and migration ability and increase apoptosis of human colorectal cancer cells,and inhibition of epithelial mesenchymal transfor?mation.
