摘 要:目的分析甲钴胺联合高压氧对急性敌敌畏中毒大鼠神经疾病靶标酯酶(NTE)、钙激活的中性蛋白酶(CANP)、缺氧诱导因子-1α(HIF-1α)mRNA表达的影响。方法SD大鼠60只随机分为4组,分别为对照组(不处理)、模型组(急性敌敌畏中毒模型)、甲钴胺组(中毒模型+甲钴胺灌胃)、甲钴胺联合HBO组(中毒模型+甲钴胺灌胃+HBO),每组15只。采用实施Y型电迷宫实验评估大鼠记忆能力。对各组大鼠血NTE蛋白,CANP蛋白含量以及脑组织HIF-1α基因表达水平进行检测。结果模型组正确反应次数>8次的大鼠比率低于对照组,差异有统计学意义(P<0.05)。甲钴胺组和甲钴胺联合HBO组大鼠正确反应次数>8次的大鼠比率显著高于模型组(P<0.05)。治疗后2周和4周,模型组血浆NTE蛋白含量显著低于对照组(P<0.05),甲钴胺组和甲钴胺联合HBO组血浆NTE蛋白含量显著高于模型组(P<0.05);治疗后2周和4周,模型组血浆CANP蛋白含量显著高于对照组(P<0.05),甲钴胺组和甲钴胺联合HBO组血浆CANP蛋白含量显著低于模型组(P<0.05),甲钴胺联合HBO组血浆CANP蛋白含量低于甲钴胺组,差异有统计学意义(P<0.05)。治疗后2周和4周,模型组脑组织HIF-1αmRNA表达量显著高于对照组(P<0.05),甲钴胺组和甲钴胺联合HBO组脑组织HIF-1αmRNA表达量显著低于模型组(P<0.05),甲钴胺联合HBO组脑组织HIF-1αmRNA表达量低于甲钴胺组,差异有统计学意义(P<0.05)。结论甲钴胺联合高压氧可较单独使用甲钴胺更有效缓解急性敌敌畏中毒大鼠智力损伤,上调NTE蛋白表达,降低大鼠CANP蛋白含量和HIF-1αmRNA表达水平。Objective To analyze the effect of Mecobalamin Combined with hyperbaric oxygen on the expression of NTE,CANP and HIF-1alpha in rats with acute dichlorvos poisoning.Methods Sixty male healthy SD rats were randomly divided into four groups,including control group(Ctrol),model group(Model),Mecobalamin group(A),Mecobalamin Combined with HBO group(B),15 rats in each group.Rats'memory ability was assessed by performing a Y-type electrical maze test.The contents of NTE protein and CANP protein in blood and the expression of HIF-1αgene in brain tissue of rats in each group were detected.Results The number of correct responses(>8 times)in the model group was lower than that in the control group,and the difference was statistically significant(P<0.05).The number of correct responses(>8 times)in the mecobalamin group and the mecobalamin combined with the HBO group was significantly higher than that in the model group(P<0.05).After 2 and 4 weeks of treatment,the plasma NTE protein content of model group was significantly lower than that of control group(P<0.05).The plasma NTE protein content of mecobalamin group and mecobalamin combined with HBO group was significantly higher than that of model group(P<0.05).At 2 and 4 weeks after treatment,the plasma CANP protein content in the model group was significantly higher than that in the control group(P<0.05).The plasma CANP protein content in the mecobalamin group and the mecobalamin combined with the HBO group was significantly lower than that in the model group(P<0.05).The HBO group was lower than that of mecobalamin group,the difference was statistically significant(P<0.05).Two and four weeks after treatment,the expression of HIF-1αmRNA in the brain tissue of the model group was significantly higher than that of the control group(P<0.05).The expression of HIF-1alpha in brain tissue of group A and B was significantly lower than that of model group(P<0.05).The expression of HIF-1αmRNA in the brain tissue of mecobalamin combined with HBO group was lower than that in the methyl
