摘 要:为明确10个马铃薯新品系在DNA水平上的遗传差异程度,用SRAP分子标记技术对其进行了比较分析。用筛选出的10对SRAP适宜引物对10个马铃薯新品系的基因组DNA扩增,得到多态性位点条带165个,多态性位点百分率占79.33%,材料间遗传差异相对较大。用特异性引物m 3/e 1建立了能明确区分出10个马铃薯新品系的SRAP指纹图。马铃薯新品系间的平均遗传距离为0.5027,以0.50为基准,将10个马铃薯材料分为四类:第一类为新品系NNCS-38、NNCS-46和NNS-07;第二类为新品系NNS-MB 9、NNS-WD 35、NNS-A 6和NNS-A 10;第三类为新品系NNS-B 12与NNS-A 1;新品系NNCS-02自成一类。In order to determine the degree of genetic difference of 10 new potato varieties at the DNA level,SRAP molecular marker technology was used for comparative analysis.The genomic DNA of 10 new potato varieties was amplified with 10 selected primers pairs of SRAP and 165 bands of polymorphic sites were obtained,with the percentage of polymorphic sites accounting for 79.33%,which reflected the relatively large genetic differences among materials.Specific primer m 3/e 1 was used to establish SRAP finger markers that could clearly distinguish 10 new potato varieties.The mean genetic distance between new potato varieties was 0.5027.Based on 0.50 genetic distance,10 potato varieties were divided into four categories:the first category was NNCS-38,NNCS-46 and NNS-07;The second was NNS-MB 9,NNS-WD 35,NNS-A 6 and NNS-A 10;And the third category was NNS-B 12 and NNS-A 1,while the new variety NNCS-02 was of its own type..
