肺癌组织中Livin、信号转导转录活化因子3基因的表达观察

摘　　要：目的观察肺癌组织中Livin、信号转导转录活化因子3(STAT3)基因的表达变化,并探讨其与临床病理特征的关系。方法肺癌患者86例,采用荧光定量PCR法检测患者肺癌组织及其癌旁组织(距离癌组织>5 cm)中Livin mRNA和STAT3 mRNA,分析Livin、STAT3基因表达与肺癌患者临床病理学特征之间的关系,采用Pearson相关分析肺癌组织中Livin mRNA与STAT3 mRNA表达的关系。结果肺癌组织中Livin mRNA、STAT3 mRNA的相对表达量分别为10.24±2.35、8.23±2.27,癌旁组织中的相对表达量分别为2.39±0.41、1.08±0.23,两组比较,P均<0.05。肺癌组织中Livin mRNA、STAT3 mRNA表达与患者肿瘤组织分化程度、淋巴结转移和肿瘤分期相关(P均<0.05),与患者的年龄、性别、吸烟史、病理分型和肿瘤体积大小无关(P均>0.05)。肺癌组织中Livin mRNA与STAT3 mRNA表达呈正相关(r=0.607,P<0.05)。结论肺癌组织中Livin mRNA和STAT3 mRNA表达上调,二者与患者肿瘤组织的分化程度、淋巴结转移和肿瘤分期密切相关,二者存在协同关系。Objective To observe the expression of Livin and signal transducer and activator of transcription 3(STAT3)gene in the lung cancer tissues and to explore their relationship with the clinicopathological features.Methods Livin mRNA and STAT3 mRNA in the lung cancer and its adjacent tissues(distance from cancer tissues>5 cm)from 86 patients with lung cancer were detected by fluorescence quantitative PCR.The relationship between the expression of Livin and STAT3 gene and clinicopathological features of lung cancer patients was analyzed.Pearson correlation was used to analyze the relationship between Livin mRNA and STAT3 mRNA expression.Results The relative expression levels of Livin mRNA and STAT3 mRNA were 10.24±2.35 and 8.23±2.27,respectively,in the lung cancer tissues,versus 2.39±0.41 and 1.08±0.23,respectively,in the adjacent tissues,with statistically significant difference between the two groups(all P<0.05).The expression of Livin mRNA and STAT3 mRNA in the lung cancer tissues were related to the degree of tumor differentiation,lymph node metastasis,and tumor stage(all P<0.05),but not to the age,gender,smoking history,pathological classification,or tumor volume(all P>0.05).Livin mRNA was positively correlated with STAT3 mRNA expression in the lung cancer tissues(r=0.607,P<0.05).Conclusion Livin mRNA and STAT3 mRNA are up-regulated in the lung cancer tissues,which are closely related to the degree of tumor differentiation,lymph node metastasis and tumor stage,and there is a synergistic relationship between them.
