摘 要:目的探讨β-榄香烯联合卡铂对结肠癌SW480细胞放疗增敏作用。方法 2019年3-5月于海南省第三人民医院肿瘤中心分子生物实验室进行实验。收集SW480细胞分为SW480细胞组(培养环境为37℃、5%CO2、20%O2)、放疗组(放射剂量为8 Gy,时间为24 h)、β-榄香烯组(80.0μg/ml)、卡铂组(100μg/ml)、β-榄香烯联合卡铂+放疗组(β-榄香烯、卡铂浓度分别为80.0μg/ml、100μg/ml,放射能量为8 Gy,时间为24 h);培养结束后,MTT法测定细胞SW480细胞增殖情况、培养计数细胞克隆形成数目,流式细胞仪测定细胞凋亡水平、细胞周期、Matrigel跨膜细胞侵袭,RT-PCR法测定SW480细胞Caspase-3、Caspase-6基因表达,Western-blot法测定SW480细胞Caspase-3、Caspase-6蛋白表达。结果与SW480细胞组比较,放疗组、β-榄香烯组、卡铂组、β-榄香烯联合卡铂+放疗组SW480细胞OD值、存活率水平、克隆形成数目降低,凋亡率、G1期比例及Caspase-3、Caspase-6 mRNA/蛋白表达水平升高(F=16.365、21.654、25.654、15.654、18.598、19.987、25.654、32.365、26.987,P均=0.000);与放疗组、β-榄香烯组、卡铂组比较,β-榄香烯联合卡铂+放疗组SW480细胞OD值、存活率水平、克隆形成数目降低,G1期比例、凋亡率及Caspase-3、Caspase-6 mRNA/蛋白表达水平升高(P<0.05)。结论β-榄香烯联合卡铂能增加结肠癌SW480细胞放疗敏感性,抑制结肠癌SW480细胞增殖、侵袭水平,提高凋亡水平,其机制与β-榄香烯联合卡铂能增促进SW480细胞Caspase-3、Caspase-6 mRNA和蛋白高表达有关。Objective To investigate the effect of β-elemene combined with carboplatin on radiosensitization of colon cancer SW480 cells.Methods From March to May 2019,experiments were performed in the Molecular Biology Laboratory of the Cancer Center of the Third People’s Hospital of Hainan Province.SW480 cells were collected and divided into SW480 cell group(culture environment:37℃,5% CO2,20% O2),radiotherapy group(radiation dose:8 Gy,time:24 h),β-elemene group(80.0 μg/ml),Carboplatin group(100 μg/ml),β-elemene combined with carboplatin+radiotherapy group(β-elemene,carboplatin concentration are 80.0 μg/ml,100 μg/ml,radiation energy is 8 Gy,time for 24 h).After the end of the culture,the proliferation of SW480 cells was measured by MTT method.The number of cell clone formation was counted in culture.Flow cytometry was used to determine the level of apoptosis,cell cycle,and invasion of Matrigel transmembrane cells.RT-PCR method was used to determine the expression of Caspase-3 and Caspase-6 in SW480 cells.Western-blot was used to determine the expression of Caspase-3 and Caspase-6 in SW480 cells.Results Compared with SW480 cell group,the radiotherapy group,β-elemene group,carboplatin group,β-elemene combined with carboplatin+radiotherapy group SW480 cell OD value,survival level,number of colony formation decreased,apoptosis rate,G1 phase ratio and Caspase-3,Caspase-6 mRNA and protein expression levels increased(F=16.365,F=21.654,F=25.654,F=15.654,F=18.598,F=19.987,F=25.654,F=32.365,F=26.987,P=0.000).Compared with the radiotherapy group,β-elemene group,and carboplatin group,β-elemene combined with carboplatin+radiotherapy group reduced the OD value,survival rate,and number of colony formation of SW480 cells,G1 phase ratio,apoptotic rate and Caspase-3,Caspase-6 mRNA and protein expression levels increased(P <0.05).Conclusion The β-elemene combined with carboplatin can increase the sensitivity of colon cancer SW480 cells to radiotherapy,inhibit the proliferation and invasion of colon cancer SW480 cells,and incr
