摘  要:目的探讨BAG-1及MCM2在子宫颈癌组织中的表达及临床意义。方法采用免疫组化EnVision两步法检测121例子宫颈癌及51例正常子宫颈组织中BAG-1及MCM2蛋白的表达;采用RT-qPCR法检测34例子宫颈癌组织及27例正常子宫颈组织中BAG-1及MCM2 mRNA的表达,并分析两者表达与子宫颈癌临床病理特征的关系。结果BAG-1在子宫颈癌及正常子宫颈组织中的高表达率为70.2%(85/121)及9.8%(5/51),两组相比差异有统计学意义(χ^2=52.545,P<0.05);MCM2在子宫颈癌及正常子宫颈组织中的高表达率为82.6%(100/121)及5.9%(3/51),两组相比差异有统计学意义(χ^2=84.836,P<0.05)。子宫颈癌组织中BAG-1及MCM2 mRNA相对表达量均高于正常子宫颈组织(P<0.05)。BAG-1高表达与FIGO分期和淋巴结转移相关(P<0.05),MCM2高表达与淋巴结转移相关(P<0.05),两者高表达与患者年龄、肿瘤直径、分化程度、病理类型及浸润深度均无关(P>0.05)。经Spearman相关分析显示,子宫颈癌中BAG-1与MCM2表达呈正相关(r=0.370,P<0.05)。BAG-1与MCM2共同高表达与FIGO分期和淋巴结转移相关(P<0.05)。结论BAG-1及MCM2可能参与了子宫颈癌的发生、发展、侵袭及转移,检测两者的表达可以作为判断子宫颈癌侵袭及预后的依据。Purpose To investigate the expression and clinical significance of BAG-1 and MCM2 in cervical cancer tissues.Methods The expression of BAG-1 and MCM2 protein in 121 cervical cancer tissues and 51 normal cervical tissues was detected by immunohistochemistry,and the expression of BAG-1 and MCM2 mRNA in 34 cervical cancer tissues and 27 normal cervical tissues was detected by RT-qPCR.The correlation between the expression of BAG-1 and MCM2 protein and the clinicopathological features was analyzed.Results The expression rate of BAG-1 in cervical cancer and normal cervical tissues was 70.2%(85/121)and 9.8%(5/51),there was a significant difference between the two groups(χ^2=52.545,P<0.05).The expression rate of MCM2 in cervical cancer and normal cervical tissues was 82.6%(100/121)and 5.9%(3/51),there was a significant difference between the two groups(χ^2=84.836,P<0.05).The relative expression of BAG-1 and MCM2 mRNA in cervical cancer tissues was higher than that in normal cervical tissues(P<0.05).High expression of BAG-1 was correlated with FIGO stage and lymph node metastasis(P<0.05),and that of MCM2 with lymph node metastasis(P<0.05).There was no correlation of high expression of BAG-1 and MCM2 with patient’s age,tumor diameter,tumor differentiation,pathological type and depth of invasion(P>0.05).There was a positive correlation between BAG-1 and MCM2 by Spearman test analysis(r=0.370,P<0.05).Co-expression of BAG-1 and MCM2 was correlated with both FIGO stage and lymph node metastasis.Conclusion BAG-1 and MCM2 may be involved in the occurrence,development,invasion and metastasis of cervical cancer.Detection of the expression of BAG-1 and MCM2 can be used as an evidence for judging the invasion and prognosis of cervical cancer.
