摘 要:主要研究了沉淀法分离提取发酵液中多粘菌素E(Colistin sulfate)的生产工艺。对超滤液热絮凝、吸附脱色、碱性沉淀以及浓缩液二次脱色等工艺参数进行了考察,结果表明:工艺最佳条件:超滤液90~95℃热絮凝5 min后,板换快速冷却,复合脱色处理:加碳量0.1%、亚硫酸盐0.5%~1.0%,沉淀条件:pH值为11.0~12.5、15~30℃、静置1 h,浓缩液脱色添加0.4%~0.8%活性炭,处理0.5 h,最高收率达到90%以上,成品各项指标符合EP生产标准。通过连续稳定性试验对各个参数进行验证,得到了适用于工业化生产的多粘菌素E分离工艺。The production process of separation and extraction of Colistin sulfate from fermentation broth by precipitation method was studied.Thermal flocculation of ultrafiltration,decolorization of activated carbon,alkaline precipitation and secondary decolorization were studied.The optimum conditions of the process:the ultrafiltrate was thermally flocculated at 90-95℃for 5 mins,then cooled and filtered.0.1%activated carbon and 0.5-1.0%sulfites were used for the first decolorization treatment.The precipitation conditions were pH 11.0-2.5 and temperature 15-25℃,and lastly precipitated for 1 hour.The concentrate was decolorized with 0.4-0.8%activated carbon for half an hour.The rate reached over 90%,and the finished products meet the EP production standards.Through the test,each parameter was verified,and the separation process suitable for industrial production was obtained.
