摘 要:目的利用Vav1-Cre介导的ROSA26R^YFP小鼠产生黄色荧光蛋白(YFP)的特性,检测其在不同器官中巨噬细胞的标记效率。方法通过基因型鉴定方法筛选出Vav1^Cre小鼠与ROSA26R^YFP小鼠杂交产生的双阳性子代小鼠,流式细胞术检测小鼠肝、脾、肾和肺中巨噬细胞的标记效率。结果Vav1^Cre与ROSA26R^YFP小鼠的杂交后代共19只,其中双阳性基因型小鼠(Vav1^CreROSA26R^YFP)9只;流式细胞术分析肝、脾、肾和肺中巨噬细胞YFP阳性细胞百分比(%)分别为91.49±1.332,98.02±0.3023,94.01±0.8127,91.39±1.47。结论Vav1^CreROSA26R^YFP小鼠能高效标记各组织器官中的巨噬细胞。Objective To detect the labeling efficiency of yellow fluorescent protein(YFP)in macrophages in different organs by using the characteristics of green fluorescent protein(GFP)produced by Vav1-Cre mediated ROSA26R^YFP mice.Methods Vav1^Cre mice and ROSA26R^YFP mice were identified by genotyping,and the YFP labeling efficiency of macrophages in the liver,spleen,kidney and lung were detected by flow cytometry.Results The percentage of YFP positive cells in the liver,spleen,kidney and lung macrophages by flow cytometry was 91.49±1.332,98.02±0.3023,94.01±0.8127 and 91.39±1.47,respectively.Conclusion Vav1^Cre strain can be efficiently recombined in macrophages(>90%)when crossing with a conditional reporter stain expressing YFP from the ubiquitously active Rosa26 locus(ROSA26RYFP).
