摘  要:罗布麻具有生态价值、药用价值和工业价值,但是核酸数据较少,阻碍了罗布麻分子生物学研究。本试验基于罗布麻转录组数据注释挖掘罗布麻中黄酮类化合物合成相关基因,为功能基因的挖掘奠定基础。本研究基于罗布麻转录组数据,通过拼接和组装,对罗布麻转录组进行了功能注释、黄酮合成相关基因的挖掘、查尔酮异构酶的克隆和分析以及SSR预测。通过对罗布麻高通量测序数据进行拼接,共获得64713条转录本,40575条Unigene,平均长度720 bp,其中28063条Unigene能被NR、GO、COG、Uniprot、PFAM和拟南芥蛋白数据库进行注释。注释表明,共有110条Unigene与罗布麻黄酮类化合物合成途径相关。本研究首次对罗布麻转录组进行了注释,鉴定出与黄酮类化合物生物合成有关的基因,为罗布麻的分子生物学研究提供了重要数据。Apocynum venetum has ecological value,medicinal value and industrial value.However,fewer nucleic acid data hinder the A.venetum molecular biology research.In this paper,based on the A.venetum data annotation results mining A.venetum flavonoids related genes,to lay the foundation for the functional gene mining.In this study,based on A.venetum transcriptome data,functional annotation of A.venetum transcriptome,mining of flavonoid-related genes,cloning and analysis of chalcone isomerase,and SSR prediction were carried out by splicing and assembly.By assembling A.venetum transcriptome sequences,64713 transcripts and 40575 Unigenes were obtained.The average length of the Unigenes was 720 bp.A total of 28063 Unigenes were annotated by aligning to the public databases NR,GO,COG,Uniprot,PFAM,and Arabidopsis Protein Database.Based on the annotation results,a total of 110 Unigenes were found to be related to the synthesis of A.flavonoids.This study was the first comprehensive transcriptome analysis in A.venetum and the genes related to flavonoids biosynthesis wereobtained,which provided important data for the molecular biology research in A.venetum.
