摘  要:转录因子Snail是胚胎发育和癌症中上皮-间充质转换的主要调节分子,可能参与调节子痫前期的发病过程。本研究通过检测Snail及其下游靶标e-cadherin在子痫前期产妇胎盘和高盐诱导的子痫前期大鼠模型胎盘中的表达,以期Snail在子痫前期发病中的作用机制。应用Western blotting检测Snail和e-cadherin在胎盘组织中的表达。通过免疫组织化学定位胎盘组织中Snail和e-cadherin的表达。研究显示,Snail蛋白在子痫前期产妇胎盘组织中明显下调了38.54%,而e-cadherin蛋白在子痫前期产妇胎盘组织中明显上调了41.75%。本研究成功建立了高盐诱导的子痫前期大鼠模型,在高盐诱导3周后,模型组妊娠大鼠均出现了子痫前期的症状。与对照妊娠大鼠相比,在高盐诱导的子痫前期大鼠胎盘中Snail蛋白表达量显著下调了55.45%,而e-cadherin蛋白水平显著上调了68.04%。Snail蛋白主要在绒毛细胞滋养层细胞和间充质细胞中表达,e-cadherin主要在绒毛基底膜和绒毛细胞滋养层细胞质中表达。提示子痫前期产妇胎盘组织中低表达的Snail导致了e-cadherin的上调,进而抑制上皮间质转化,降低了滋养细胞浸润能力,导致子痫前期产妇的胎盘发育异常。The transcription factor Snail is a major regulator of epithelial-mesenchymal transition in embryonic development and cancer and may be involved in the regulation of preeclampsia.This study examined the expression of Snail and its downstream target e-cadherin in the placenta of preeclampsia delivery women and high salt-induced preeclampsia rat models,in order to predict the mechanism of Snail in the pathogenesis of preeclampsia.Western blotting was used to detect the expression of Snail and e-cadherin in placenta.The expression of Snail and e-cadherin in placental tissue was localized by immunohistochemistry.Studies have shown that Snail protein was significantly down-regulated by 38.54%in tissue of preeclampsia delivery women,while e-cadherin protein was significantly up-regulated by 41.75%in tissue of preeclampsia delivery women.In this study,a rat model of pre-eclampsia induced by high salt was successfully established.After 3 weeks of high salt induction,the pregnant rats in the model group developed symptoms of pre-eclampsia.Compared with control pregnant rats,the expression of Snail protein in the placenta of rats with high salt-induced preeclampsia was significantly down-regulated by 55.45%,while the level of e-cadherin protein was significantly up-regulated by 68.04%.Snail protein was mainly expressed in chorionic trophoblast cells and mesenchymal cells,and e-cadherin was mainly expressed in the cytoplasm of villus basal membrane and chorionic trophoblast.It was suggested that the low expression of Snail in the preeclampsia maternal placenta tissue leads to the up-regulation of e-cadherin,which inhibits epithelial-mesenchymal transition and reduces the infiltration ability of trophoblasts,leading to abnormal placental development in preeclampsia.
