摘 要:目的:研究人参皂苷20(S)-Rh2联合BMAP28肽对肺癌细胞A549凋亡的影响及作用机制。方法:实验分为对照组、人参皂苷20(S)-Rh2组、BMAP28肽组和联合用药组[20(S)-Rh2联合BMAP28肽]。CCK-8试剂盒检测各组A549细胞增殖情况;DAPI/FITC-BMAP28荧光双染法检测各组肺癌A549细胞凋亡形态学变化;流式细胞术检测各组肺癌A549细胞凋亡率。Western blot检测PARP、Caspase-9、Cytochrome C和Bax蛋白的表达变化。结果:人参皂苷20(S)-Rh2、BMAP28肽及联合用药对肺癌A549细胞的增殖均有抑制作用,联合用药组的增殖抑制作用具有协同效应。荧光双染法可见联合用药组细胞凋亡形态变化明显。BMAP28肽和联合用药组可以促使PARP(Caspase-3底物)和Caspase-9裂解;人参皂苷20(S)-Rh2、BMAP28肽及联合用药组上调Cytochrome C和Bax蛋白表达,联合用药组效果更加显著。结论:人参皂苷20(S)-Rh2和BMAP28肽具有协同作用,两者通过线粒体信号通路抑制肺癌A549细胞增殖,诱导其凋亡。Objective:To study the effect and mechanism of ginsenoside 20(S)-Rh2 combined with BMAP28 peptide on apoptosis of lung cancer A549 cells.Methods:The experimental setup was divided into four groups:Control group(A549),ginsenoside 20(S)-Rh2 group,BMAP28 peptide group and combination group[20(S)-Rh2 combined BMAP28 peptide].CCK-8 kit was used to detect the proliferation of lung cancer A549 cells in each group.DAPI/FITC-BMAP 28 fluorescence double staining method was used to detect the morphological changes of apoptosis in lung cancer A549 cells of each group.Flow cytometry was used to detect the apoptosis rate of lung cancer A549 cells in each group.Results:The Ginsenoside 20(S)-Rh2 group,BMAP28 peptide group and the combination group had inhibitory effect on the proliferation of lung cancer A549 cells,and the combination group had synergistic effect.The morphological changes of apoptosis were observed by fluorescence double staining.BMAP28 peptide group and combination group could induce the cleavage of PARP and Caspase-9.Cytochrome C and Bax protein expression were up-regulated in ginsenoside 20(S)-Rh2 group,BMAP28 peptide group and combination group,the effect was more significant in the combination group.Conclusion:The Ginsenoside 20(S)-Rh2 and BMAP28 peptide have a synergistic effect,both of which inhibit the proliferation and induce the apoptosis of lung cancer A549 cells through the mitochondrial signaling pathway.
