摘 要:目的观察二氢杨梅素(DHM)对人膀胱癌细胞增殖的影响,并探讨其分子机制.方法用不同浓度(0、5、20μmol/L)DHM处理UMUC3细胞,通过噻唑蓝(MTT)实验和流式细胞术检测DHM对细胞增殖的抑制作用,实时荧光定量聚合酶链反应(FQ-PCR)和蛋白质印迹法(Westem blot)检测DHM对周期蛋白(Cyclin D1和Cyclin E1)以及周期蛋白依赖性激酶(CDK2和CDK4)的影响.用SPSS 21.0软件进行统计分析,结果采用均值±标准差(Mean±SD)表示,统计方法为单因素方差分析,多组间两两比较采用最小显著性差异(LSD)法和邓肯(Duncan)多重范围检验.结果DHM明显抑制UMUC3细胞增殖.与对照组[(48.4±0.6)%]比较,低浓度组[(55.2±0.1)%]和高浓度组[(60.5±0.3)%]G0/G1期细胞比例显著上升,差异有统计学意义(低浓度组F=417.668,P<0.01,高浓度组F=401.652,P<0.01).高浓度组G0/G1期细胞比例大于低浓度组,差异有统计学意义(F=100.221,P<0.05).RT-PCR和Western blot结果显示Cyclin D1、Cyclin E1、CDK2、CDK4表达均明显降低.结论DHM可抑制人膀胱癌细胞增殖,可能与阻滞细胞周期由G0/G1期向S期过渡有关.Objective To evaluate the effect and mechanism of dihydromyricetin(DHM)on the proliferation of human bladder cancer cells.Methods UMUC3 cells were incubated with DHM at the concentrations of 0,5 and 20 pimol/L respectively,then the inhibitory effect were investigated by thiazole blue(MTT)assay and flow cytometry.The expression of Cyclin DI and Cyclin El and cyclin-dependent kinases(CDK2 and CDK4)was detected using real-time fluorescence quantitativepolymerase chain reaction(FQ-PCR)and Western blotting.Statistical analysis was performed using SPSS 21.0 software and the results were expressed by mean value土standard deviation(Mean±SD).The statistical method was one-way analysis of variance incorporated with the least significant difference(LSD)method and Duncan multiple range test.Results DHM inhibited the proliferation of UMUC3 cells obviously.Compared with the control group[(48.4±0.6)%],the percentage of G0/Gx phase cells in low concentration group[(55.2±0.1)%]and high concentration group[(60.5±0.3)%]increased,and the difference was statistically significant(F=417.668 in the low concentration group,P<0.01,F=401.652 in the high concentration group,P<0.01).The percentage of Go/G]phase cells was higher in the high concentration group than in the low concentration group,and the difference was statistically significant(F=100.221,P<0.05).The results of RT-PCR and Western blotting showed that the expression of CDK2,CDK4,Cyclin DI and Cyclin El decreased obviously.Conclusion DHM can effectively inhibit the proliferation of human bladder cancer cells,and its mechanism may be related to blocking the transition of cell cycle from Go/G|phase to S phase.
