摘 要:目的探讨环指蛋白43(RNF43)在人类乳腺癌组织中的表达及其意义.方法利用实时定量反转录聚合酶链反应(RT-qPCR)法、蛋白质印迹法(Western blot)检测60例乳腺癌组织及相应的60例癌旁正常组织中RNF43的mRNA、蛋白表达水平并进行统计学分析.结果RNF43mRNA及蛋白表达在乳腺癌组织的表达量(分别为1.216±0.112、0.439±0.056)显著高于癌旁正常乳腺组织(0.253±0.018、0.107±0.015)(P<0.05).RNF43 mRNA及蛋白表达在浸润性乳腺癌淋巴结转移组(分别为1.713±0.021、0.611±0.026)高于无淋巴结转移组(0.648±0.019、0.242±0.017),瘤周脉管肿瘤侵犯组(1.681±0.009、0.584±0.062)高于无侵犯组(0.860±0.014、0.328±0.071),TNM分期(Ⅲ期,1.713±0.022、0.637±0.014)高于TNM分期(Ⅰ~Ⅱ期,0.928±0.041、0.324±0.050),组织学分级(Ⅱ~Ⅲ级,1.305±0.053、0.471±0.019)高于组织学分级(Ⅰ级,0.638±0.044、0.231±0.071),雌激素受体(ER)表达阳性组(1.443±0.077、0.525±0.017)高于ER表达阴性组(0.726±0.033、0.253±0.007),孕激素受体(PR)表达阳性组(1.439±0.071、0.519±0.020)高于PR表达阴性组(0.857±0.069、0.310±0.010),细胞核增殖抗原(Ki-67)高阳性率组(≥30%,1.401±0.039、0.519±0.014)高于低阳性率组(<30%,0.661±0.071、0.286±0.006),差异均有统计学意义(P<0.05).RNF43 mRNA及蛋白表达与患者的年龄(<35岁组1.124±0.017、0.419±0.037;≥35岁组1.282±0.024、0.453±0.006)、肿瘤大小(≤2 cm组,1.100±0.051、0.412±0.023;>2 cm组,1.262±0.014、0.450±0.030)、病理类型(浸润性导管癌组,1.243±0.079、0.448±0.011;浸润性小叶癌组,1.108±0.044、0.403±0.013)、人表皮生长因子受体2(Her-2)表达(阳性组,1.178±0.009、0.422±0.017;阴性组,1.230±0.011、0.445±0.010)及月经状态(绝经前,1.231±0.049、0.440±0.008;绝经后,1.167±0.056、0.436±0.011)均无明显相关(P>0.05).结论RNF43 mRNA及其蛋白的异常表达可能与乳腺癌的发生、发展、侵袭及转移有关.Objective To investigate the expression and significance of ring finger protein 43(RNF43)in human breast cancer tissue.Methods Real-time quantitative reverse transcriptase-polymerase chain reaction(RT-qPCR)and Western blotting were used to detect the mRNA and protein expression levels of RNF43 in breast cancertissues(60 cases)and normaladjacent tissues(60 cases).Results The expression of RNF43 mRNA and protein in breast cancer tissues(1.216+0.112,0.439±0.056)was significandy higher than that in adjacent normal breast tissues(0.253±0.018,0.107±0.015)(P<0.05).RNF43 mRNA and protein expression in node-positive invasive breast cancer(1.713±0.021,0.611±0.026)was higher thannode-negative(0.648±0.019,0.242±0.017),peritumoral vascular invasion group(1.681±0.009,0.584±0.062)higher than the non-invasive group(0.860±0.014,0.328±0.071),TNM staging(stage HI,1.713±0.022,0.637±0.014)was higher than TNM staging(stage I-U,0.928±0.041,0.324±0.050),histological grade(U-HI,1.305±0.053,0.471±0.019)was higher than histological grade(1,0.638±0.044,0.231±0.071),and estrogen receptor(ER)expression positive group(1.443±0.077,0.525±0.017),The expression was higher than the ER expression negative group(0.726±0.033,0.253±0.007),and the progesterone receptor(PR)expression positive group(1.439±0.071,0.519±0.020)was higher than the PR expression negative group(0.857±0.069,0.310±0.010),the high proliferation cell nuclear antigen(Ki-67)positive rate group(M30%,1.401±0.039,0.519±0.014)was higher than the low positive rate group(<30%,0.661±0.071,0.286±0.006),and the difference was statistically significant(P<0.05).RNF43 mRNA and protein expression was not related to patientsage(<35 years old group:1.124±0.017,0.419±0.037;M35 years old group:1.282±0.024,0.453±0.006),tumor size(W2 cm group:1.100±0.051,0.412±0.023;>2 cm group:1.262±0.014,0.450±0.030),pathological type(invasive ductal carcinoma group:1.243±0.079,0.448±0.011;invasive lobular carcinoma group:1.108±0.044,0.403±0.013),human epidermalgrowth factor
